摘　　要：目的 :观察异搏定 (verapamil ,Ver)对非p -gp高表达细胞株的增敏及耐药逆转作用。方法 :使用二甲氧唑黄比色法 (XTT法 )检测药物敏感性 ,观察Ver对鬼臼乙叉甙 (etoposide ,VP16)的增敏及耐药逆转作用。结果 :单独Ver对HL 60和HL 60 /Adr细胞株的IC50 分别为 2 6.13mg·L-1和 3 9.0 0mg·L-1,单独VP 16对HL 60细胞的IC50 为 2 47.17μg·L-1,而加用Ver 5mg·L-1预处理 1h后 ,VP16对HL 60细胞的IC50 值下降为原来的 1/1.73 ;单独VP 16对HL -60 /Adr细胞的IC50 为872 .97μg·L-1,而加用Ver 5mg·L-1预处理 1h后 ,IC50 下降至 2 0 8.45 μg·L-1,为原来的 1/4.19,与VP16对HL 60敏感株的IC50 接近。结论 :Ver对非pObjective The experiment was designed to observe the chemosensitive and reversal effect of verapamil( Ver) on non?p?gp overexpressed cells.Method 2,3 bis(2 methoxy 4 nitro 5 sulfophenyl) 5 〔(phenylamino)Carbonyl〕 2H tetrazolium hydroxide(XTT) bioassay was used.Results IC 50 of verapamil alone to HL?60 and HL?60/Adr cells was about 26.13 mg·L -1 and 39.00?mg·L -1 ,respectively.IC 50 of etoposide(VP16) alone to HL?60 and HL?60/Adr cells was about 247.17?μg·L -1 and 872.97?μg·L -1 which were reduced to 1/1.73 and 1/4.19,respectively,when incubated with Ver 5?mg·L -1 .Conclusion Ver has a chemosensitive and reversal effect on non p gp overexpressed cells.
