摘 要:目的 :探讨大鼠烧伤休克时CD11a及CD11b单克隆抗体 (CD11aMAb、CD11bMAb)对白细胞与内皮细胞粘附的影响及对重症烧伤休克大鼠的作用。方法 :在细胞液流室中 ,观察CD11aMAb及CD11bMAb处理的重症烧伤休克大鼠白细胞与培养的鼠肺血管内皮细胞间粘附性在不同切应力下的变化 ;复制大鼠重症烧伤休克模型 ,颈动脉插管给予单克隆抗体 ,观察动物存活时间的变化。结果 :与对照组相比CD11bMAb可显著降低重症烧伤休克大鼠白细胞的粘附率 (P <0 0 5 ) ,明显延长动物存活时间 (P <0 0 5 )。而CD11aMAb则无明显效果 (P >0 0 5 )。结论 :CD11bMAb能降低白细胞对血管内皮细胞的粘附 ,延长重症烧伤休克大鼠存活时间。Objective:To investigate the influence of CD11a and CD11b monoclonal antibody(CD11aMAb, CD11bMAb) on leukocyte-endothelial cells (ECs) adhesion and the effects on severely burnt shock rats. Methods:The changes of adhesion between the leukocytes isolated from the severely burnt shock rats which were treated with CD11aMAb or CD11bMAb and the cultured vascular ECs from the rats' lungs were observed under the different shearing stresses in a cell flow chamber. The model of severely burnt shock rats were replicated and CD11aMAb or CD11bMAb was administered through the carotid artery cannulas, and the changes of their survival time were observed. Results:Compared with the control group, the CD11bMAb could significantly reduce the leukocyte adhesion rate of the severely burnt shock rats ( P <0 05), and the survival time was remarkably prolonged ( P <0 05), but the CD11aMAb did not show the significant effect ( P >0 05). Conclusion:The CD11bMAb can reduce the adhesion between the leukocytes and the ECs and prolong the survival time of the severely burnt shock rats.
