摘 要:目的观察三七总皂苷(PNS)对DN的肾脏保护作用及机制。方法 36只雄性SD大鼠分成正常组、模型组、PNS干预组(均12只)。4,8周末,检测血清和肾皮质的晚期糖基化终产物(AGEs)、超氧化物歧化酶(SOD)、丙二醛(MDA)以及肾皮质VEGF含量。结果 PNS干预组,血清与肾组织的MDA、AGEs和VEGF含量,均明显低于模型组(P<0.01,P<0.05);而SOD显著高于模型组(P<0.05)。结论 PNS可能通过抗氧化、抑制AGEs生成、下调肾组织VEGF表达,来延缓DN大鼠肾脏病变进展。Objective To explore the protective effect of panax notoginseng saponins(PNS) in rat with diabetic nephropathy(DN).Methods A total of 36 model rats were randomly divided into normal group(n =12),model group(n = 12) and PNS treatment group(n=12).The rat DN model was induced by intraperitoneal injection of streptozotocin in the left lower abdomen of the rats.In the experiment the 4th weekend and 8th weekend,serum and renal cortex supperoxide dismutase(SOD) and malondialdehyde(MDA) of the rats in each group were measured by colorimetric assay.Serum and renal tissue advanced glycation end products(AGEs) were determined by spectrum law of fluorescence.The expression of VEGF in renal tissue was observed by the immunohistochemistry.Results In the diabetic animals treated with PNS,AGEs and MDA in serum and renal tissue and VEGF of renal tissue were lower than those in diabetic animals(P〈0.01 or P〈0.05),and the SOD in serum and renal tissue were increased obviously(P〈0.05).Conclusion PNS can slow down renal disease progression in DN rats through anti-oxidant,inhibiting overproduction of AGEs and downregulating the expression of VEGF in renal tissue.
