槲皮素对人结肠癌SW480细胞体外侵袭能力的影响

摘　　要：目的:研究槲皮素对结肠癌SW480细胞的体外侵袭能力的影响,并探讨其可能的作用机制。方法:结肠癌SW480细胞经槲皮素处理后用Transwell小室的方法检测其体外黏附、侵袭和运动能力。明胶酶谱分析法检测SW480细胞分泌的基质金属蛋白酶活性。结果:结肠癌SW480细胞经槲皮素处理后,体外侵袭、运动能力呈剂量依赖性下降,SW480细胞MMP-2及MMP-9的分泌下降(P<0.05)。结论:槲皮素在体外剂量依赖性地抑制结肠癌SW480细胞的转移能力,这可能与槲皮素抑制MMP-2及MMP-9的分泌有关。Objective：To observe the effect on the invasion of human colon cancer SW480 cell line in vitro induced by quercetin.Methods： Human colon cancer cells SW480 were treated with different doses of quercetin,and the invasion of SW480 was determined by Transwell chamber,the expressions of MMP-2 and MMP-9 were detected by gelatin zymography.Results： Transmembrane cell and the expressions of MMP-2 and MMP-9 of quercetin group decreased in dose-dependent manners（P0.05）.Conclusion： Quercetin can inhibit invasion of human colon cancer cells,and decreasing the expression of MMP-2 and MMP-9 may play an important role in its mechanism.
