摘 要:目的观察一氧化氮(NO)信号通路及胃泌素(GAS)水平在大鼠脾气虚证进程中的变化及益气健脾中药的干预效应。方法将受试动物随机分为正常组、模型组(7、14、21 d组)、益气健脾组,每组10只。采用大黄法、力竭法及饥饿法复合建立脾气虚证大鼠模型,分时段测定各组大鼠胃肠组织、血清一氧化氮合酶(NOS)、NO、GAS动态表达水平。结果与正常对照组比较,模型大鼠胃肠组织及血清NOS、NO、GAS水平逐渐降低(P<0.05,P<0.01),且以21 d组显著,而益气健脾组大鼠NOS、NO、GAS水平未见明显变化,且与模型21 d组比较差异有统计学意义(P<0.05)。结论 NO、NOS、GAS在脾气虚的形成过程中主要以低水平表达为主,而益气健脾中药具有促进NO信号通路及GAS表达的作用。Objective To investigate the dynamic changes of nitric oxide signal pathway and gastrin(GAS) expression in spleen-qi deficiency rats and effect of Yiqi Jianpi herbs.Methods Rats were randomly divided into 5 groups:normal control group,model group(observed on 7,14 and 21 d),and Yiqi Jianpi herb group,10 rats in each group.The spleen qi deficiency model rats were made by rhubarb,exhaustive and hungry method,and treatment group was treated with Yiqi Jianpi herbs.Nitric oxide signal pathway and GAS expression of rats were evaluated on 7,14 and 21 d.Results In model group,levels of NOS,NO and GAS were much lower than that of normal control group(P0.05,P0.01),and was obvious in 21 d group.NOS,NO and GAS of Yiqi Jianpi herb group had no significant changes,and were much higher than that of model 21 d group(P0.05).Conclusion The nitric oxide signal pathway and GAS expression in spleen-qi deficiency are mainly low-level expression and Yiqi Jianpi herbs has the function of elevating the expression of NOS,NO and GAS.
