摘  要:目的观察抗纤灵药物血清对骨髓来源的成纤维细胞转化生长因子-β(TGF-β)和Ⅰ型胶原(collagenⅠ)的抑制作用。方法将抗纤灵方煎至含原药材3.2 g/mL,福辛普利配成含药0.33 mg/mL,给大鼠灌胃(正常组给予蒸馏水灌胃),制备抗纤灵血清、福辛普利血清和正常血清。用DMEM培养基稀释血清,将其分为正常血清组、福辛普利组、抗纤灵组、TGF-β1组、TGF-β1+福辛普利组和TGF-β1+抗纤灵组。以大鼠肾组织体外培养骨髓来源的成纤维细胞为材料,采用酶联免疫吸附法和实时荧光定量PCR法检测骨髓来源的成纤维细胞collagenⅠ和TGF-β的表达。结果加入TGF-β1刺激因子的细胞表达TGF-β和CollagenⅠ较正常大鼠血清组明显增强,抗纤灵及福辛普利药物血清可明显抑制其表达。结论抗纤灵药物血清可以通过抑制骨髓来源的成纤维细胞TGF-β的表达,减少细胞外基质collagenⅠ的沉积,从而延缓肾纤维化的进程。Objective To observe the effect of Kangxianling on transforming growth factor-β(TGF-β) and collagenⅠ of bone marrow-derived fibroblast.Methods 3.2 g/mL Kangxianling and 0.33 mg/mL fosinopril was administered respectively to rats by gavage(distilled water to normal group) to prepare Kangxianling serum,fosinopril serum and normal serum.The serum was diluted with DMEM medium and divided into normal serum group,fosinopril group,Kangxianling group,TGF-β1 group,TGF-β1+fosinopril group and TGF-β1+Kangxianling group.Bone marrow-derived fibroblast was dissociated and harvested by trypsin,which was isolated from the normal rat kidney tissue.The expression of TGF-β and collagenⅠ of bone marrow-derived fibroblast were examined by enzyme-labeled immunosorbent assay and real time fluorescent quantitation polymerase chain reaction.Results TGF-β and collagenⅠ of bone marrow-derived fibroblast with TGF-β1 stimulating factor increased markedly than those in normal serum group,and were inhibited by Kangxianling and fosinopril.Conclusion Kangxianling could depress the expression of TGF-β,reduce collagenⅠ production of extracellular matrix,thereby delaying the process of renal fibrosis.
