摘 要:目的建立马兰药材中大黄酸、大黄素及大黄酚含量的测定方法。方法采用高效液相色谱法(highperformance liquid chromatography,HPLC)。色谱柱:Kromasil-C18柱(250mm×4.6mm,5μm);流动相:甲醇-0.1%磷酸水溶液;梯度洗脱;检测波长:254nm;流速:1ml/min;柱温:30℃。结果大黄酸、大黄素、大黄酚浓度分别在0.253~2.530μg/ml(r=0.999 6)、0.249~2.490μg/ml(r=0.999 3)、0.257~2.570μg/ml(r=0.999 7)范围内与峰面积呈良好的线性关系;平均加样回收率(n=6)分别为99.27%(RSD=1.28%)、99.78%(RSD=1.05%)、99.92%(RSD=0.83%)。结论所建立的方法准确、可靠、简便,适合马兰药材中大黄酸、大黄素及大黄酚含量的测定。Objective To establish a method for determining the contents of rhein, emodin, and chry- sophanol in Kalirneris indica by high performance liquid chromatography (HPLC). Methods HPLC was performed on Kromasil C18 column (250mm×4.6mm,5μm) with a mobile phase of methanol-0.1G phos-phoric acid for gradient elution at a detection wavelength of 254 nm,a flow rate of 1 ml/min,and a column temperature of 30℃. Results The concentrations of rhein,emodin,and chrysophanol showed a good linear relationship with peak areas in 0. 253-2. 530 μg/ml (r=0. 9996), 0. 249-2. 490 μg/ml (r=0. 9993), and 0. 257 2. 570μg/ml (r=0. 9997), and the average recovery rates (n=6) were 99.27% (RSD=1. 28%) 99. 78%o (RSD= 1.05 %), and 99.92%(RSD= 0.83%), respectively. Conclusion HPLC determination method can be used for measuring the contents of rhein, emodin, and chrysophanol in Kalimeri,s indica for its accuracy, reliability, and simplicity.
