摘 要:目的以贝母素甲、贝母素乙作为指标成分,研究不同浸泡时间对浙贝母有效成分提取的影响,考察比较合理的煎煮方法。方法采用高效液相色谱-蒸发光散射器(HPLC-ELSD)法检测,Dionex Acclaim C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%二乙胺(70∶30),流速为1.0 mL/min;ELSD条件为漂移管温度42℃,载气(高纯N2)流速1.0 L/min。结果不同浸泡时间对浙贝母饮片有效成分提取量差异明显。随着饮片浸泡时间的延长,煎煮后药渣的白心率降低,而煎膏得率及其有效成分提取量均增高。结论浙贝母饮片用常规煎煮方法很难煮透,至少应先浸泡1 h,才能使有效成分较好地溶出,发挥更好的药效。Objective To study the effects of different soaking time on the extraction of active components of Fritillary bulb with peimine and peiminine as index components and to investigate more reasonable decocting method. Methods The procedure of HPLC- ELSD analysis was performed on the Dionex Acclaim C18 chromatography column (250mm×4.6mm,5μm) with acetonitrile-0.1% diethylamine (70:30) as the mobile phase and the flow rate was 1.0 mL/min. The ELSD conditions were the drift tube temperature of 42 ℃ and the flow rate of carrier gas (high purity N2) of 1. 0 L/rain. Results The difference among the extracting amounts of the active components in different soaking time was obvious. With the extension of soaking time, the white heart rate of herb residue af- ter boiling was decreased, while the extract yield rate and the extracting amounts of the active components all were increased. Conclusion The decoction pieces of Fritillary bulb are difficult to be decocted thoroughly by the conventional boiling method, which should be soaked for an hour at least to make better dissolution of the active components for playing a better efficacy.
