摘 要:目的:建立连翘酯苷脂质体包封率的测定方法。方法:通过逆相蒸发法制备连翘酯苷脂质体,运用超滤离心法测定其包封率。结果:超滤离心法能快速有效的对脂质体与游离药物进行分离,游离药物平均回收率在98.75%~101.71%之间,超滤加样回收率在95.67%~98.76%之间;此法测定3批样品,所得包封率为72.77%~74.07%,RSD为0.56%~1.77%。结论:超滤离心法操作简单、快速,可用于连翘酯苷脂质体包封率的测定。Objective: To establish a method for determining entrapment efficiency of forsythoside liposomes. Methods: Forsythoside liposomes were prepared by a reverse evaporation method, and then the entrapment efficiency was determined by ultrafihration. Results: The free drug was separated from the forsythoside liposomes efficiently. Average recovery rate of free forsythoside was 98.75% - 101.71% , and the recovery rate was 95.67% - 98.76% in the process of ultrafihration. The entrapment efficiency of three lot samples was 72.77% - 74.07% with RSD of 0.56% -1.77% by ultrafihration. Conclusion: The ultrafiltration method can simply and immediately measure entrapment efficiency of forsythoside liposomes after preparation.
