三叶青提取物对肺癌细胞株A549的影响

摘　　要：目的观察三叶青提取物对肺癌A549细胞的影响并探讨其作用机制。方法体外培养A549细胞,用不同浓度(10-1g.L-1,10-2g.L-1,10-3g.L-1)的三叶青提取物处理,流式细胞术检测细胞凋亡率,westernblot法检测细胞中caspase-3的表达。结果 (1)10-1g.L-1、10-2g.L-1三叶青提取物处理A549细胞后,细胞凋亡率明显高于对照组,差异有统计学意义(P<0.05)。(2)10-1g.L-1、10-2g.L-1、10-3g.L-1三叶青提取物处理后的A549细胞内,Caspase-3蛋白的表达水平均较对照组显著升高,并随着三叶青提取物作用浓度的增高而逐渐上升,差异均有统计学意义(P<0.05)。结论三叶青提取物可促进A549细胞的凋亡,可能与其上调细胞内Caspase-3的表达有关。Objective To study the effect of extract of radix tetrastigma hemsleyani on lung cancer cell line A549 and its mechanisms. Methods A549 ceils were cultured in vitro and treated with different concentrations （10^-1g·L^-1,10^-2g·L^-1,10^-3g·L^-1） of extract of Radix Tetrastigma I-Iemsleyani. The apoptosis rate was detected by flow cytometry, and the expression of caspase-3 was detected by western blot. Results （1） Compared with the control group, the apoptosis rates ofA549 cells treated by 10^-1g·L^-1,10^-2g·L^-1 extract of P, adix Tetrastigma Hemsleyani both significantly increased （P〈0.05）. （2） Compared with the control group, the protein expression of caspase-3 in cells treated by 10^-1g·L^-1,10^-2g·L^-1 or 10^-3g·L^-1 extract of Radix Tetrastigma Hemsleyani all increased markedly （P 〈 0.05）, and it was significantly raised with the increase of concentration of extract of Radix Tetrastigrna Hemsleyani （P〈0.05）. Conclusion The extract of Radix Tetrastigrna Hemsleyani could promote the apoptosis of A549 cells, which may be related to the increase ofcaspase-3 expression.
