摘 要:本文建立了高效液相色谱法快速准确测定A549细胞内外齐墩果酸(OA)的含量。该法采用Hypersil ODS色谱柱(250×4.6mm,5μm);流动相为pH=3.38的甲醇-水(体积比为90∶10),流速1.0mL/min,柱温30℃,检测波长210nm,进样量为20μL。结果表明,细胞内液中OA在1.020~20.40μg/mL范围内线性关系良好(r=0.9996),平均回收率为97.57%;细胞外液中OA在5.100~81.60μg/mL范围内线性关系良好(r=0.9998),平均回收率为97.37%。该方法简便易行,准确度高,重现性好,可用于A549细胞内外OA浓度的动态变化规律研究。A high performance liquid chromatography method for the determination of intracellular and extracellular oleanolic acid(OA) was developed in this paper. The chromatographic conditions were as follow, stationary phase,ODS(250×4.6 ram, 5μm) . mobile phase 90 : 10 (V/V) methanol-water (pH - 3.38);flow rate 1.0 mL/min;column temperature 30 ℃ ;detection wavelength 210 nm and the injection volume 20 μL. The results indicated that the linear range of intracellular OA was 1. 020-20.40 μg/mL (r=0. 9996), the average recovery was 97.57%;and the linear range of extracellular OA was 5. 100- 81.60 μg/mL(r= 0. 9998), the average recovery was 97. 37%. The method was simple, accurate and precise,which can be used for the dynamic study of intracellular and extracellular OA.
