摘  要:目的观察安宫牛黄丸对大鼠急性缺血后神经元凋亡及神经元内磷酸化Akt表达的影响。方法 50只雄性SD大鼠随机分为假手术组、模型组、安宫牛黄丸小、中、大剂量组。采用大脑中动脉线栓法建立脑缺血模型,采用Nissl染色观察脑损组织伤,采用NeuN和TUNEL荧光双染检测神经元凋亡,NeuN和磷酸化Akt免疫荧光双染检测神经元内磷酸化Akt的表达。结果大、中剂量安宫牛黄丸可显著改善神经功能,减轻缺血引起的脑组织损伤,减少引起的神经元凋亡比率,上调神经元内磷酸化Akt的表达。结论安宫牛黄丸可能通过上调缺血后神经元内磷酸化Akt的表达,抑制神经元凋亡的发生。AIM To investigate the effects of Angong Niuhuang Pills on neuronal apoptosis and expression of phosphorylated Akt after the rats suffered from the acute ischemia.METHODS Fifty male SD rats randomized into sham,ischemia-reperfusion,and low,middle,high dose of Angong Niuhuang Pills groups.The cerebral ischemia was induced by middle cerebral artery acclusion.The neuronal apoptosis was detected by NeuN and TUNEL double stain,and the expression of phosphorylated Akt in neuron was detected by NeuN and phosphorylated Akt double stain.RESULTS Treatment with middle and high dose of Angong Ninhuang Pills improved nerological function,attenuated cerebral injury,decreased neuronal appotosis and up-regulated expression of phosphorylated Akt in neuron.CONCLUSION Angong Niuhuang Pills inhibit the neuronal apoptosis by up-regulating expression of phosphorylated Akt in neuron following cerebral ischemia.
