中国广西人群Nogo-B基因内含子区遗传多态性的检测

摘　　要：目的研究Nogo-B基因内含子区单核苷酸多态性(SNP)各等位基因及基因型在中国广西地区人群中的分布频率,比较其在不同种族间分布的差异。方法采用单碱基延伸的PCR技术和DNA测序法检测199例中国广西人的Nogo-B基因内含子区rs12464595 C/T、rs2864052 G/A多态性,并结合人类基因组计划(Hapmap)公布的四个人群(欧洲人、非洲人、日本人和中国北京人)的SNP分型数据,比较分析这五个人群的基因型及等位基因的分布频率。结果在中国广西人群中存在Nogo-B基因内含子区rs12464595 C/T、rs2864052 G/A多态性,与欧洲及非洲人群比较,Nogo-B基因多态性的分布频率均存在显著性差异(P<0.05);而与日本人及北京人比较差异无显著性(P>0.05)。结论在中国广西人群中存在Nogo-B基因多态性,但与其他种族人群比较存在显著性差异,这种差异对于人类学的研究可能起重要的作用。【Objective】To study the frequencies of allele and genotype distribution of neurite growth inhibitor-A（Nogo-B） gene 3＇-untranslated regions single nucleotide polymorphism（SNP） in Chinese Guangxi population,and to analyze the distributions of Nogo-B polymorphism among different races.【Methods】The Nogo-B gene 3＇ untranslated regions rs12464595 C/T and rs2864052 G/A polymorphisms were examined by the polymerase chain reaction-single base extension（PCR-SBE） technique and DNA sequencing methods in 199 Chinese Guangxi population,frequencies of allele and genotype of Nogo-B gene rs12464595 C/T,rs2864052 G/A polymorphisms were analyzed for Guangxi populations compared with other the four populations（HapMap-CEU,HapMap-YRI,HapMapJPT and HapMap-HCB） from Human Genome Project group（Hapmap） data.【Results】There are Nogo-B gene polymorphisms in Guangxi population.The frequencies of allele and genotype distribution of Nogo-B gene rs12464595 C/T,rs2864052 G/A polymorphisms were significant different from HapMap-CEU and HapMap-YRI population（P0.05）,the Nogo-B gene polymorphisms were not significant different compared with HapMap-JPT and HapMap-HCB（P0.05）.【Conclusion】There are Nogo-B gene polymorphisms in Guangxi population,the frequencies of polymorphism distribution of Nogo-B gene are significantly different compared with others ethnic populations.It might play an important role for anthropological research.
