摘 要:采用生物素-磁珠富集法,构建短毛切梢小蠹微卫星富集文库。在该文库中随机挑选100个克隆作测序分析,结果表明:具微卫星位点的克隆占91%;具有10次以上碱基重复次数的微卫星序列占36%,其中最高碱基重复次数为63次;在这些微卫星序列中,完美型占47.3%,非完美型占24.2%,混合型占16.5%。表明获得的微卫星文库是高质量的文库,完美型的比例高于不完美型和混合型,与真核生物微卫星序列特征相符。The enriched microsatellite library of Tomicus brevipilosus (Eggers) was constructed using biotin-magnetic bead enrichment method. One hundred clones randomly chosen from the library were sequenced. The results showed that 91% of microsateUite loci were obtained. Among which the microsatellite sequences with more than 10 times of alkaline base repetitions accounted for 36%, and the highest alkaline base repetition was 63 times. From these microsatellites, 47.3% (about 43 repeat motifs) were perfect, 24.2% (about 22 repeat motifs) were imperfect, and 16.5% (about 15 repeat motifs) were compound. The results indicated that the enriched microsatellite library was of high-quality, in which the proportion of perfect microsatellites types was higher than that of the imperfect and compound types, which was in accordance with the feature of microsatellite sequences of other eukaryote.
