摘 要:目的探讨硫酸铍(BeSO4.4H2O)对小鼠肝线粒体功能的影响。方法将36只6周龄昆明(KM)小鼠随机分为对照组、低剂量组及高剂量组3组,每组12只,雌雄各半。对照组采用灭菌生理氯化钠溶液按0.01 ml/g(体质量)隔天腹腔注射,低、高剂量组分别以1、2 mg/kg BeSO4.4H2O生理氯化钠溶液隔天腹腔注射染毒,连续染毒3周。取肝脏采用差速离心法制备线粒体,采用荧光分光光度法测定其活性氧(ROS)水平及线粒体膜电位(Δψm),采用分光光度法测定细胞色素C(Cytc)水平及线粒体肿胀程度。结果与对照组比较,低、高剂量组肝脏系数增高,肝脏线粒体ROS及Cytc水平随染毒剂量的增加而升高,差异均有统计学意义(P<0.05);随染毒剂量的增加Δψm下降、线粒体肿胀明显。结论 BeSO4.4H2O可致小鼠肝线粒体功能障碍。Objective To explore the effect of beryllium sulfate (BeSO4· 4H2O) on mice liver mitochondrial tunctinn. Methods 36 KM mice were randomly divided into 3 groups and administered with saline solution of beryllium sulfate ( l and 2 mg/kg) by intraperitoneal injection for 3 weeks. Liver mitochondria were isolated by differential centlifugation, then the level of mitoehondrial reactive oxygen species (ROS) and mitochondrial cytochrome c (Cytc) , mitoehondrial swelling and the mitochon- drial membrane potential (AtOm) were detected. Results The relative organ weight of liver in exposure groups were significantly higher than those of the controls (P 〈 0. 05 ). Compared with those nf the control group, ROS and Cyte in all exposed groups were significantly higher ( P 〈 0. 05 ). A dramatic mitochondrial swelling and a decrease of △ψm in the dose groups were found. Conclusion Beryllium sulfate can induce mitochondrial dysfunction on mice liver mitochondrial.
