摘  要:目的观察β榄香烯对转化生长因子(TGF-β)刺激后的肾小球系膜细胞(HMC)表达结缔组织生长因子(CTGF)及纤维连接蛋白(FN)的影响。方法先用有效浓度10 ng/mL的转化生长因子(TGF-β)刺激肾小球系膜细胞(HMC)24 h,然后再用不同浓度,分别为80μg/mL、120μg/mL、160μg/mL的榄香烯干预被刺激的肾小球系膜细胞(HMC)24 h。实验分高中低不同浓度组、诱导组及空白对照组。用Western蛋白印迹法测定FN蛋白表达和CTGF的蛋白表达。用RT-PCR法检测FN和CTGF的mRNA水平的变化。结果经TGF-β刺激24 h后的肾小球系膜细胞(HMC),诱导组较空白组比较,FN蛋白和CTGF蛋白的表达能明显(P<0.05),高浓度β榄香烯组和中浓度β榄香烯组较诱导组比较,HMC上的FN蛋白和CTGF蛋白的表达和mRNA水平明显降低(P<0.05),并呈浓度依赖方式降低MHC上表达的FN和CTGF的表达。结论β榄香烯可以通过减少诱导纤维化后的肾小球系膜细胞中CTGF和FN的表达,发挥抑制肾间质纤维化的作用,β榄香烯可以抑制肾间质纤维化,在延缓肾功能衰竭过程中起着重要作用。【Objective】 Observe the effect of β-elemene on expression of CTGF(Connective Tissue Growth Factor) and FN(Fibronectin) on human renal membrane cells(HMC) that stimulated by TGF-β.【Methods】 HMC is stimulated by TGF-β of the concentration of 10μg/mL for 24 h.After 24 h,the human renal membrane cells(HMC) were treated with the β-elemene of different concentrations respectively as 80μg/mL,120μg/mL and 160μg/mL for 24 h.CTGF and FN protein expression were detected by Western Blot.RT-PCR was performed on renal tissue to detect FN and CTF mRNA expression.【Results】 CTGF and FN expression on HMC stimulated by TGF-β were raised evidently,compared to the blank group.CTGF and FN expression level on HMC,which were intervened by β-elemene,were less than the induction group.It reduced the expression of CTGF and FN level in a dose dependent manner.【Conclusion】 β-elemene has the effect of inhibiting the tubulointerstitial fibrosis by the path of restraining CTGF and FN expression.It suggests that β-elemene can inhibit the tubulointerstitial fibrosis and retarder progression of CDK.
