IL-4及其受体和IL-13基因多态性与儿童哮喘的相关性研究

摘　　要：目的探讨IL-4基因启动子C589T,IL-4R基因E375A、C406R、Q576R和IL-13基因Arg110Gln位点多态性与儿童哮喘发生的相关性。方法采用聚合酶链反应-限制性片段长度多态性分析(PCR-PFLP)方法检测122名哮喘儿童、144名健康儿童各个多态性位点的基因型,采用SPSS 13.0进行统计学分析。结果IL-4R基因E375A、C406R这两个位点的基因型及等位基因频率分布在哮喘组与正常对照组均存在显著性差异(P<0.05),哮喘组E375A位点等位基因A频率显著高于正常对照组;C406R位点的等位基因C频率分布在哮喘组显著高于正常对照组。IL-13基因Arg110Gln位点Gln等位基因携带者(GlnGln+ArgGln)的频率在哮喘组显著高于对照组。结论 IL-4R基因E375A和C406R位点可能与儿童哮喘的发病存在关系,其中,A和C等位基因可能分别是易感等位基因,而IL-13基因Gln携带者可能更易罹患哮喘。【Objective】 To investigate the relationship between the C589T of interleukin-4,E375A,C406R,Q576R of interleukin-4 receptor and Arg110Gln of interleukin-13 gene SNPs polymorphisms and Childhood asthma.【Methods】 Genomic DNA was isolated from the venous blood leukocytes of 122 unrelated patients with Childhood asthma and 144 healthy unrelated individuals（control group）.Polymorphism C589T,E375A,C406R,Q576R and Arg110Gln were genotyped by PCR-restriction fragment length polymorphisms（PCR-RFLP）.Genotype and allele frequencies were analyzed by SPSS13.0 software.【Results】 There were significant differences for both allele and genotype frequencies of E375A and C406R of IL-4R gene between the Childhood asthma and control group.The allele A of E375A and C allele of C406R in childhood asthma group was significantly higher than that in control group（P 0.05）,the genotype frequencies of Gln carriers（GlnGln＋ArgGln） were significantly higher in childhood asthma compared to control（P 0.05）.【Conclusion】 There is an association between E375A,C406R in the IL-4 and ArgGln in the IL-13 gene and childhood asthma.The individuals with A allele of E375A and C allele of C406r are susceptible to childhood asthma.The Gln carriers are also disposition to childhood asthma.
