摘  要:利用mRNA差异显示 (DifferentialDisplayReverseTranscriptPCR ,简称DDRT PCR)技术比较了正常供和缺铁胁迫下铁高效型小麦 (京— 4 11)的铁低效型小麦 (三属麦— 3)的基因表达差异模式 .此差异模式暗示出两个品种表型差异的遗传背景 .利用挑选出的差异显示cDNA片段为探针进行Northern杂交 ,证明一些差异cDNA的表达受缺铁胁迫的抑制 .测序和序列分析结果表明 ,受抑制的cDNA片段分别与Cytc基因、ATP binding transporters基因、和dDTP 4 ,6 葡萄糖脱水酶的基因有一定同源性 .Expression difference of gene under iron\|deficiency and iron\|efficiency were compared between iron\|efficient wheat cultivar Jing—411 and iron\|inefficient wheat cultivar Sanshumai—3 by differential display Reverse Transcript PCR (DDRT\|PCR) method. Northern blotting were carried out with root tips of wheat cultival Jing—411 which treated by iron\|deficiency stress and iron\|efficiency, and the probes come from the special cDNA fragment selected from DDRT\|PCR products of the same cultival. The results demonstrated some cDNAs expression were suppressed in iron\|deficiency condition. Sequened analysis showed that cDNA fragment were homogenous to Cytochrome C gene\,ATP\|binding transporters (ABC transporters) gene and dTDP\|D\|glucose 4,6\|dehydratase.
