摘 要:目的:研究绞股蓝总皂苷纳米乳的制备及其抗衰老作用。方法:通过对绞股蓝总皂苷溶解度试验和伪三元相图确定其最佳处方并制备出绞股蓝总皂苷纳米乳,用透射电镜和粒度分析仪分析其形态和对粒径进行鉴定。以皮下注射D-半乳糖复制亚急性衰老大鼠模型。30只SD大鼠均分为6组,即正常对照(2mL生理盐水)、亚急性衰老模型(2mL生理盐水)、绞股蓝水溶液(120mg.kg-1)和绞股蓝总皂苷纳米乳高、中、低剂量(180、120、60mg.kg-1)组;25只老年大鼠均分为5组,即自然衰老模型(2mL生理盐水)、绞股蓝水溶液(120mg.kg-1)和绞股蓝纳米乳高、中、低剂量(180、120、60mg.kg-1)组。灌胃给药,每天1次,连续30d。检测大鼠血清、下丘脑和心脏的丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、一氧化氮(NO)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)水平。结果:绞股蓝总皂苷纳米乳最佳处方为聚氧乙烯氢化蓖麻油(RH40)/肉豆蔻酸异丙酮(IPM)/丙二醇/水,表面活性剂和助活性剂比例为3:1,平均粒径为35.8nm。与亚急性衰老模型和自然衰老模型组比较,绞股蓝总皂苷纳米乳高、中、低剂量组大鼠血清、心脏和下丘脑组织中的SOD、CAT、T-AOC、GSH-Px水平显著升高(P<0.05),而NO、MDA含量显著减少(P<0.05)。结论:绞股蓝总皂苷纳米乳有较好的抗衰老作用,且呈剂量依赖关系。OBJECTIVE:To study preparation and anti-aging effects of gypenosides nanoemulsion.METHODS:The optimal prescription was determined by solubility test and pseudo-ternary phase diagram of gypenosides nanoemulsion,and then gypenosides nanoemulsion was prepared.The morphology and particle size of nanoemulsion were analyzed by transmission electron microscope and particle size analyzer.The subacute aging rat model was induced by D-galactose.The rats were divided into 6 groups,such as normal control group(2 mL physiological saline),subacute aging model group(2 mL physiological saline),gynostemmapenta phylum solution group(120 mg.kg-1)and gypenosides nanoemulsion high-dose,middle-dose and low-dose groups(180,120,60 mg.kg-1).Twenty-five aged rats were divided into 5 groups,such as aged model group,gynostemmapenta phylum solution group(120 mg.kg-1) and gypenosides nanoemulsion high-dose,middle-dose and low-dose groups(180,120,60 mg.kg-1).They were given relevant medicine intragastrically once a day for consecutive 30 days.MDA,SOD,CAT,NO,GSH-Px,total antioxidant capacity(T-AOC) were all detected in serum,hypothalamus and heart.RESULTS:The optimal prescription of gypenosides nanoemulsion was as follows:RH40,IPM.propylene glycol,water(Km=3:1),average diameter of 35.8 nm.Compared with acute aging model group and aged model group,the levels of SOD,CAT,T-AOC and GSH-Px in serum,hypothalamus and heart of gypenosides nanoemulsion high-dose,middle-dose and low-dose groups increased significantly(P〈0.05),while the contents of NO and MDA decreased significantly(P〈0.05).CONCLUSION:Gypenosides nanoemulsion is good against aging,in a dose dependent manner.
