摘 要:目的:制备蛇床子素β-环糊精包合物,并考察其稳定性。方法:采用逆向混合搅拌法制备蛇床子素β-环糊精包合物,用紫外分光光度法测定包合物中蛇床子素的含量、溶解度、溶出度;以蛇床子素含量为考察指标,对蛇床子素β-环糊精包合物进行强光照射、高温和高湿试验。结果:经显微镜观察、紫外光谱、热分析等方法证明包合物确已形成,包合物的溶解度约是原药的34倍,120min时包合物的累积溶出量约是原药的2.5倍。在光、热、湿等因素影响下,包合物中蛇床子素含量没有明显变化,而蛇床子素与β-环糊精物理混合物的含量明显下降。结论:包合物能显著改善蛇床子素的溶解度和溶出度,蛇床子素β-环糊精包合物具有较好的抗光照性、热稳定性和湿稳定性,其稳定性明显优于蛇床子素混合物。OBJECTIVE:To prepare the inclusion complex of osthol-β-cyclodextrin(osthol-β-CD),and to investigate the stability of inclusion complex.METHODS:The inclusion complex of osthol-β-CD was prepared by counter mixture stirring method.The content,solubility and dissolution of osthol in inclusion complex were determined by UV spectrophotometry.Using the content of osthol as the inspection index,the inclusion complex of osthol-β-CD was examined with intense light,high temperature and humidity test.RESULTS:The formation of the inclusion complex was confirmed by microscope examination,UV absorption spectrum scanning,thermal analysis,etc.The solubility of inclusion complex increased about 34 times.The accumulative dissolving quantity of inclusion complex was about 2.5 times the drug substance within 120 min.The content of osthol showed no significant changes in the inclusion compounds but noticeable changes in the physical mixture of osthol and β-cyclodextrin.CONCLUSION:The results indicate that the inclusion complex can improve the solubility and dissolution of osthol.Osthol-β-CD has the characteristics of anti-light,anti-heat and anti-humidity,and the stability of it is better than that of osthol mixture
