摘 要:应用流式细胞仪观察了不同光暗交替周期对莱哈衣藻的配子形成过程影响 ,并采用核酸抑制剂和蛋白合成抑制剂处理 ,观察其配子配对情况。结果表明在缺氮诱导条件下 ,前期的光照对配子分化形成十分重要 ,而先黑暗后光照或连续黑暗诱导时 ,细胞或配子的分裂缺乏规律性。光暗周期处理有利于配子的同步发育 ,光照期对雄性配子发育特别重要。同在缺氮条件下 ,不同的光暗周期处理的配子配对状况不同 ,在正常光暗交替周期处理时 ,配对的百分率高 ,而连续光照或先黑暗再光照则配对率低 ,连续黑暗时配对率为 0。当用核酸和蛋白合成抑制剂处理时 ,配对率显著降低。其中核酸抑制剂的作用主要在前期 ,而蛋白合成抑制剂的抑制时间更长。Observed the effect of various light/dark system on gameto genesis in C. reinhardi by flow cytomtry, then observed that effect of 5FU a nd CHM on gametic fusion. The results showed, when cultural condition of nitrogen stress early light in induction was significant for gametogenesis, but in early dark or successive dark induction rule of cell mitosis or gametogenesis was diso rdered. Normal period of light/dark benefited development of gametes. Light prof ited to development of male gamete. In condition of nitrogen stress, when induct ion of normal light/dark period percentage of magetic fusion was the highest, ho wever in condition of continuous light or early dark ratio of gametic fusion was less. In condition of continuous dark gametic fusion was not observed. Ratio of magetic fusion was signification inhibited by 5FU and CHM. Inhibition of CHM wa s longer tim than that of 5FU. The results were discussed.
