摘  要:目的:探讨清心开窍方及其皂苷成分对Aβ25-35杏仁核注射诱导AD大鼠学习记忆能力及脑组织凋亡信号转导分子,Aβ及βAPP表达的影响。方法:雄性SD大鼠40只,随机分为正常组、模型组、安理申组、清心开窍方组和皂苷组,每组8只。采用双侧杏仁核注射Aβ25-35(10 g.L-1)制造AD大鼠模型,造模后第2天起,分别灌胃给药安理申(1.67 mg.kg-1)、清心开窍方水煎剂(12.67 mL.kg-1)、皂苷(6.30 mg.kg-1)及双蒸水,2周后,观察大鼠水迷宫空间记忆能力,并通过免疫组化方法研究脑组织Caspase-3,Aβ及βAPP的表达。结果:中药各组能明显改善AD大鼠学习记忆能力,与对照组比较有显著性差异(P<0.05,P<0.01);清心开窍方组、皂苷组大鼠大脑皮层、海马区Caspase-3,Aβ及βAPP表达水平降低,与模型组比较差异非常显著(P<0.01,P<0.05)。结论:清心开窍方及其皂苷能显著改善AD大鼠学习记忆的能力,可能是通过降低皮质、海马Caspase-3,Aβ及βAPP表达发挥作用。Abstract:Objective: To study the effect of Qingxin Kaiqiao formula and saponin on the learning and memory ability and the expression of the apoptosis signal transducers Aβ and βAPP in AD rat brain. Method: The comparative observation method was adopted for the animal test. Forty male SD rats were randomly divided into five groups, namely the normal group, the model group, the aricept group, the Qingxin Kaiqiao formula group and the saponin group, with eight rats in each group. Aβ25-35 (10 g·L -1) was injected into their bilateral amygdala to establish the AD rat model. Since the next day, they were intragastrically administered with Aricept (1.67 mg·kg-1), Qingxin Kaiqiao decoction (12.67 mL·kg-1), saponin (6.30 mg·kg-1) and double distilled water filling for 2 weeks to observe their spatial memory ability in a Morris water maze and study the expression of Caspase-3, Aβ and βAPP in brain tissues by immunohistochemistry. Result: Each traditional Chinese medicine groups showed significant improvement in the learning and memory ability of AD rats and notable differences (P〈0.05,P〈0.01) compared with the control group. The Qingxin Kaiqiao formula group and the saponin group showed a decrease in the expressions of Caspase-3, Aβ and βAPP in cerebral cortex and hippocampus area, displaying notable differences (P〈0.01,P〈0.05) compared with the control group. Conclusion: Qingxin Kaiqiao formula and saponin can obviously improve the learning and memory ability of AD rats with by decreasing the expression of Caspase-3, Aβ and βAPP in cortex and hippocampus.
