摘 要:目的:建立不同产地野菊花中绿原酸、3,4-二咖啡酰奎尼酸、3,5-二咖啡酰奎尼酸的含量测定。方法:采用HPLC方法,色谱柱为Diamonsil(4.6 mm×250 mm,5μm),甲醇-0.1%磷酸溶液做流动相进行梯度洗脱,检测波长326 nm,柱温25℃,流速0.8 mL.min-1。结果:绿原酸、3,4-二咖啡酰奎尼酸和3,5-二咖啡酰奎尼酸的线性范围分别为1.00~20.00,4.64~92.80,2.233~44.66 mg.L-1;加样回收率分别为98.97%,99.14%;99.39%,RSD分别为1.81%,1.66%,1.36%(n=6)。结论:方法操作简便,分析速度快,结果准确,重复性好,可用于野菊花中绿原酸、3,4-二咖啡酰奎尼酸、3,5-二咖啡酰奎尼酸的含量测定。Objective: To determine the content of chlorogenic acid, 3, 4-O-dicaffeoylquinic acid, 3, 5-O-dieaffeoylquinic acid in Chrysanthemi Indici Flos of different areas. Method: HPLC was used. The chromatographic separation was performed on a Diamonsil CIs (4.6 mm ~ 250 mm, 5 p^m) column eluted with gradient mobile phase of methanol-O. 1% Ha PO4, the detection wavelength was set at 326 nm and column temperature was set at 25 ~C. Result: Nice linear relation between the area and content when the amount was within 1.00-20. 00, 4.64-92. 80, 2. 233-44.66 mg ~ L-1 The recoveries were 98.97% , 99. 14% and 99.39% respectively, RSD was 1.81% , 1.66% and 1.36% (n =6) respectively. Coneluslon: The method is accurate, reproducible and suitable to determine these active components in Chrysanthemi Indici Flos. It can be used for quality control of Chrysanthemi Indici Flos.
