摘  要:目的:阐明跨膜丝氨酸蛋白酶6(TMPRSS6)在低氧预防运动性低血色素中对机体铁状态的调节机制,为预防运动性低血色素发生提供理论基础。方法:24只雄性Wistar大鼠随机分为安静对照组(CG)、运动性低血色素组(EG)和运动后低氧恢复组(HE),分别用ELISA检测了血清铁代谢指标,RT-PCR检测肝铁代谢相关基因表达。结果:1)HE组Hb、SF、骨髓铁显著高于EG(Hb:HE:139.56±10.24g/L,EG:113.16±17.36g/L,P<0.01;SF:HE:29.72±2.85ng/mL,EG:18.23±3.57ng/mL,P<0.01;骨髓铁:HE:75.98±7.08μg/g,EG:62.19±8.98μg/g,P<0.05)。2)HE组肝HIF-1和TMPRSS6mRNA显著高于EG(P<0.05)。3)HE组m-HJV和hepcidin mRNA和肝铁显著低于EG(肝铁含量:HE:80.18±13.66μg/g,EG:111.51±12.64μg/g,P<0.01)。结论:从组织层面和分子生物学层面上进一步支持和完善了低氧对铁代谢的调控模型:通过HIF-1刺激TMPRSS6高表达,降低hepcidin,从而促进肝释铁蛋白FPN1表达,贮存铁释放,满足运动中机体合成Hb对铁的需求,这可能是防止运动性低血色素发生的原因之一。Objective;To illuminate effects of hypoxia on iron metabolism in the prevention of exercise-associated anemia. Methods: 24 male Wistar rats were randomly divided into three groups, the control group (CG), and the strenuously exercised group (EG), and the hypoxia exercise group (13.6% O2,8h/d, HE). The content of erythropoietin (EPO) and serum Fer- ritin (SF) were examined by ELISA kits (Abcam). The mRNAs of hepatic hepcidin, hypoxia- inducible factor 1 (HIF-1), transmembrane serine proteases 6 (TMPRSS6) and hemoiuvelin (m-HJV) were examined by RT-PCR. Results: 1)Compared with EG, the Hb, SF, and iron storage in bone marrow of HE were significantly increased(Hb: HE: 139. 56 ± 10. 24 g/L, EG: 113.16±17.36 g/L,P〈0.01;SF.-HE.-29.72±2.85 ng/mL,EG.. 18. 234-3.57 ng/ mL,P〈0.01 iron storage in bone marrow; HE: 75.98 7.08 μg/g, EG: 62. 19±8.98 /μg/ g,P〈0.05). 2)Compared with EG, the mRNAs of HIF-land TMPRSS6 in the liver of HE were significantly increased. 3)The mRNAs of HJV and Hepcidin of HE were decreased compared to EG, and the iron storage in liver was decreased in HE (HE; 80. 18±13. 66 μg/g, EG..111.51± 12. 64 μg/g, P〈 0. 01 ). Conclusions:Moderate hypoxia plays an important role in preventing exercise-induced iron deficiency. Exposure to hypoxia, the increased HIF-1 in liver stimulates the expression of TMPRSS6, and as a result, the synthesis of Hepcidin is decreased, which leads to the iron mobilization in liver to meet the requirement of iron during strenuous exercise,which prevents the exercise-associated anemia.
