摘  要:[目的]建立资木瓜多糖的提取和含量测定方法。[方法]采用乙醇回流提取法提取资木瓜中的多糖,并利用苯酚-硫酸法测定多糖的含量。[结果]线性回归方程为:Y=0.008 8X-0.010 9(R2=0.992 3)。试验结果表明,在1.0~5.0μg/ml范围内,葡萄糖浓度与吸光度的线性关系良好。提取2次后,脱脂木瓜中多糖的含量为13.21%;未脱脂木瓜中多糖的含量为9.26%;平均回收率为97.04%,RSD为3.52%。[结论]该方法简便易行,稳定性、精密度和重现性良好,可用于资木瓜中多糖的提取和含量测定。[Objective] To establish a method for the extraction and determination of polysaccharide from Chaenomeles speciosa.[Method] Polysaccharides were extracted by the ethanol reflux method from Chaenomeles speciosa,and then determined by the method of C6H5OH phenol-H2SO4.[Result] The linear regression equation was Y=0.008 8X-0.010 9(R2=0.992 3).As indicated by the results,the glucose concentration showed good linear relations with the absorbency within the range of 1.0-5.0 μg/ml.As twice of extraction,the content of polysaccharides was about 13.21% in skimmed Chaenomeles speciosa,and 9.26% in non-skimmed Chaenomeles speciosa.The average recovery was 97.04% with RSD of 3.52%.[Conclusion] This method is simple with high stability,precision and reproducibility,which can be used for the extraction and determination of polysaccharides from Chaenomeles speciosa.
