摘 要:在Plackett-Burman试验设计基础上,以温度、pH和CaCO3添加量为自变量,通过Design-Expert中心组合实验,建立培养条件优化的二次回归模型,再通过对OD600(1/50)的响应曲面分析,确定蜡样芽孢杆菌培养的最佳工艺条件。由响应面分析结果可知:在温度为34.36℃,pH为7.37,CaCO3添加量为2.4 g/L最优条件下,预测最大OD600(1/50)平均值为0.652。采用上述优化培养条件进行5组平行试验,OD600(1/50)的平均值为0.649,表明实际试验值与模型预测值基本一致。实验结果表明,采用基于中心组合设计的响应面分析方法能够较大地提高蜡样芽孢杆菌的产率,培养液中活菌数可高达2.48×1010个/mL。The optimum conditions of cultivation of Bacillus cereus were studied by the response surface analysis to the OD600(1/50).Based on the Plackett-Burman experimental design,the independent variables of the temperature,pH,and CaCO3 dosage were determined,and a quadratic regression model of optimized culture conditions was established by Design-Expert center combination experiments.The results of response surface analysis showed that temperature of 34.36 ℃,pH of 7.37,CaCO3 dosage of 2.4 g/L were the optimal conditions.The predicted maximal OD600 was(1/50) average of 0.652.A set of parallel tests was made under optimal culture conditions,and the average OD600(1/50) was 0.649,indicating that the actual experimental values and model prediction ones were basically the same.Results showed that the response surface methodology using central composite design could improve the yield of Bacillus cereus,the number of viable cells in the culture medium could be 2.48×1010 cfu/mL.
