摘  要:目的观察pU-VEGF-siRNA真核表达载体转染骨髓间质干细胞后血管内皮生长因子(VEGF)mRNA表达及VEGF蛋白质的变化。方法脂质体介导pU-VEGF-siRNA质粒转染体外培养的人骨髓间质干细胞,RT-PCR方法检测转染后骨髓间质干细胞VEGF mRNA表达水平,免疫印迹法半定量检测转染后骨髓间质干细胞VEGF蛋白质水平。结果荧光显微镜提示脂质体介导质粒转染人骨髓间质干细胞成功,且效率较高;与对照组相比,VEGF mRNA及蛋白质表达水平在pU-VEGF-siRNA转染组显著降低,空载体转染组无明显变化。结论 pU-VEGF-siRNA质粒转染骨髓间质干细胞,可降低骨髓间质干细胞VEGF mRNA表达水平,下调其VEGF蛋白质水平。Objective To investigate the expressions of vascular endothelial growth factor (VEGF) mRNA and protein in bone mes- enchymal stem cells (BMSCs) transfected with pU-VEGF-siRNA. Methods pU-VEGF-siRNA was transfected into BMSCs by liposome. Expression of VEGF mRNA in BMSCs was detected by RT-PCR. Expression of VEGF protein in BMSCs was detected by Western blot. Re- suits pU-VEGF-siRNA was transfected into BMSCs by liposome successfully and the efficiency was high. Compared with control group, ex- pressions of VEGF mRNA and protein were lower in transfected group and had no change in empty vector transfected group. Conclusions Transfecting pU-VEGF-siRNA into BMSCs could decrease the expressions of VEGF mRNA and protein.
