甲状旁腺激素对大鼠绝经后骨质疏松牙周炎模型外周血中成纤维细胞生长因子23与碱性磷酸酶表达的影响

摘　　要：目的:通过动物实验,检测大鼠外周血中成纤维细胞生长因子23(FGF23)与碱性磷酸酶(ALP)水平的变化,探讨甲状旁腺激素(PTH)对其表达的影响及其在治疗牙周炎过程中的机理。方法:选用68只3月龄雌性Wistar大鼠随机分为健康对照组(A组),伪手术对照组(B组),去势牙周炎组(C组),去势牙周炎并甲状旁腺激素注射组(D组)。C组和D组去势牙周炎模型建立成功后,D组间歇性低量腹腔内注射甲状旁腺激素,A、B和C组腹腔内注射等体积的生理盐水。两个月后,麻醉下方块截取包括实验区磨牙的双侧上颌骨,HE染色后进行观察。收集实验各阶段外周血,检测FGF23、ALP水平。结果:注射PTH后,D组外周血FGF23、ALP水平高于A、B、C组(P<0.05),D组牙槽骨中骨小梁较C组粗大、致密,排列有规则,与A、B组相似。结论:甲状旁腺激素可升高外周血FGF23、ALP的水平,有利于成骨,对绝经期后牙周炎有治疗作用。Objective： Detection of the level of fibroblast factor 23 （FGF23） and alkaline phosphatase （ALP） in the rat peripheral blood through animal experiments to explore the affect of Parathyroid Hormone （PTH） to the expression of these two factors and the mechanism of in the couse of the treatment of periodontitis. Method： 68 3monthold female Wistar rats were randomly divided into the healthy control group （Group A）, sham control group （Group B）, castration periodontitis （Group C）, castration periodontitis and parathyroid hormone injection group （Group D）.After Group C and Group D castra tion periodontitis model was successfully established, Group D inject parathyroid hormone in the abdominal cavity by inter mittent and low volume, Group A, B and C inject the same of saline in the abdominal cavity.After two months, box intercep tion of the bilateral maxillary including molar in the experimental zone, and observed after HE staining, detecting the level of FGF23 and ALP through the collection of peripheral blood. Result： The level of FGF23,ALP in peripheral blood of Group D was higher than group A,B,C （P 〈O.05）.The trabecular bone of alveolar bone of Group D are thicker, denser and more regularly arranged than that of Group C, similar to the Group A and B. Conclusion： Parathyroid hormone can increase the level of FGF23 and ALP in peripheral blood, and conducive to osteogenesis.h has therapeutic effect on Postmenopausal peri odontitis.
