电针足三里和昆仑对佐剂性关节炎大鼠的治疗作用

摘　　要：目的:建立佐剂性类风湿关节炎大鼠模型,研究电针刺激"足三里"和"昆仑"穴对佐剂性关节炎大鼠的治疗作用,为针灸治疗风湿性关节炎提供理论依据。方法:在大鼠右后足掌面由掌腱膜向踝关节处皮下注入0.1 mL弗氏完全佐剂,诱发佐剂性关节炎大鼠模型。并用0.25 mm×40 mm针灸针刺入大鼠一侧"足三里"和"昆仑"穴,深度为5 mm,并连接HANS电针仪进行电针治疗,调节电针仪频率2 Hz,强度2 mA,每天1次,每次30min,连续针灸28d后,观察不同组大鼠关节炎指数和一般状态并测量足趾肿胀度和踝关节病理形态学变化。结果:大鼠注射弗氏完全佐剂后,足跖肿胀度比空白组明显增加,说明已成功建立佐剂性类风湿关节类大鼠模型。电针治疗14d以内,治疗组和模型组大鼠足跖肿胀度无显著差异,但治疗28d后,治疗组大鼠足跖肿胀度和关节症状评分降低,足爪组织炎症情况有所减轻。此外,大鼠注射弗氏完全佐剂及电针治疗不会影响大鼠正常发育。结论:电针刺激"足三里"和"昆仑"穴对佐剂性关节炎具有较好的疗效。The rat model of adjuvant arthritis was established and the therapeutic effect of electroacupuncture in ＂Zusanli＂ and ＂Kunlun＂ holes of adjuvant arthritis rats was studied for providing a theoretical basis for acupuncture treatment of rheumatoid arthritis. Methods： The rat model of adjuvant arthritis was established with injection of 0.1mL Freund＇s complete adjuvant in right hind Pall, and stick 0.25 mm×40 mm acupuncture needle into the side of ＂Zusanli＂ and ＂Kunlun＂ hole at 5 mm in depth, Then connected the HANS instrument for electroacupuncture treatment.The treatment last for 28d with frequency for 2 Hz and intensity for 2 m.A at one time per day and each time for 30min.The arthritis index and toe swelling were measured every week and the pathological changes of ankle were observed atter treatment for 28d. Results： Toe swelling of rats injected with Freund＇s complete adjuvant was more serious than the normal rats, illustrated the successful estabilishment of adjuvant arthritis rats model. After treatment with electroacupuncture for 14d, there was no difference in rats paw swelling between the treated group and model group. But after 28di the rat toe swelling and joint symptoms score decreased, while toe tissue inflammation eased in the rats of treatment group compared with the model group during the same period. In addition, injection of Freund＇s complete adjuvant and electroacupuncture treatment would not affect the normal development of rats. Conclusions： Stimulation with electric needle in ＂Zusanli＂ and ＂Kunlun＂ hole have a good curative effect on adjuvant arthritis.
