摘  要:目的探讨苯甲酸雌二醇对去卵巢大鼠皮肤中TGFβ1/Smad3信号通路蛋白表达的影响。方法35只健康雌性Wistar大鼠被随机分为4组:A组,正常对照组(n=5);B组,假手术组(n=10);C组,去卵巢组(即手术组,n=10);D组,去卵巢苯甲酸雌二醇治疗组(即治疗组,n=10)。各组按要求制备动物模型。运用免疫组化法检测各组大鼠皮肤组织中TGFβ1、Smad3的变化,用平均光密度值表示,半定量分析结果。结果与A组相比,D组大鼠皮肤组织中的TGFβ1及Smad3表达明显升高(P〈0.05);B组TGFβ1及Smad3的表达无明显变化(P〉0.05);C组TGFβ1及smad3的表达明显降低(P〈0.05)。结论苯甲酸雌二醇通过上调皮肤组织中TGFβ1、Smad3分子表达,增强TGFβ1/Smad3信号通路功能而发挥生物学效应。Objective To investigate the effect of estradiol benzoate on the protein expression of TGFβ1/Smad3 signal transduction pathway in the skin of ovariectomized rats. Methods Thirty-five female Wistar rats were randomly divided into four groups. Group A was normal control group ( n = 5 ). Group B was sham-ovariectomy ( n= 10). Group C was ovariectomy ( n= 10). Group D was ovariectomy and estradiol ben- zoate replacement ( n= 10). Animal model was made as planning. The expression of TGFβ1 and Smad3 in the skin tissue of Wistar rats was determined by immunohistochemical staining with specific antibody. Mean optic density was analyzed. Results The expression of TGFβ1 and Smad3 in group D was statistically higher than that in group A ( P 〈 0.05 ). And the expression of TGF[31 and Smad3 in group B was also the same as that in group A ( P 〉 0.05 ), while the expression of TGFβ1 and Smad3 in group C was statistically lower than that in group A ( P 〈 0.05). Conclusion Estradiol benzoate can directly regulate the fibroblast proliferation in the skin tissue of rats through affecting the expression of TGFβ1 and Smad3 in the skin tissue, which may be relat- ed to postponing skin aging.
