摘  要:目的:构建羊膜上皮细胞(AECs)胶原海绵复合体,并研究其细胞生长及特性。方法:将大鼠AECs接种于胶原海绵支架中,一般贴壁培养细胞作对照,用CM-Dil及Hoechest33342标记细胞膜和细胞核、HE染色、扫描电镜等观察细胞的生长和形态,CCK-8法检测细胞活力,免疫细胞化学、实时荧光定量PCR检测细胞表型。结果:与一般贴壁培养AECs相比,AECs在胶原海绵中粘附生长良好,增殖能力强,直径变小,细胞表型变化不明显,仅NestinmRNA表达上调。结论:胶原海绵支架与AECs生物相容性良好,能维持细胞原特性并促进其生长和增殖。Objective: To construct complex of amniotic epithelial ceils (AECs) and collagen sponge and to investigate its cel- lular characteristics. Methods: Rat AECs were seeded in the scaffold of collagen sponge and in the bottle for attachment cul- ture served as a control group. The growth and morphological changes were observed with CM-Dil labeling cell membrane, Hoechest33342 labeling nucleus, HE staining, scanning electron microscopy. Cellular viability was detected with CCK-8 kit. Cellular phenotype was examined by immunohistochemistry and real time quantitative PCR. Results: Compared with the culture in the bottle, rat AECs adhered and grew well within the scaffold, proliferated more actively. Its diameter de- creased. Its cellular phenotype did not change obviously, but nestin mRNA upregulated. Conclusion: Collagen sponge and AECs is proven to have a good biocompatibility, which could maintain previous cellular characteristics of AECs and promote AECs growth and proliferation.
