摘　　要：目的：检测Oct-4和生殖细胞核因子（GCNF）在小鼠精原干细胞（SSCs）诱导分化前后的表达。方法：体外分离培养小鼠SSCs，用干细胞因子（SCF）诱导其向精母细胞方向分化，通过间接免疫荧光显色和RT-PCR，检测Oct-4和GCNF在小鼠SSCs诱导分化前后的表达。结果：Oct-4在小鼠SaCs诱导前有表达，诱导2d后表达下降；而GCNF在小鼠SaCs诱导前呈阴性表达，向精母细胞诱导2d后呈阳性表达。结论：GCNF可能通过抑制Oct-4的表达促使SSCs分化。Objective： To detect the expression of Oct-4 and germ cell nuclear factors （GCNF） before and after spermatogoni- um stem cells （SSCs） are induced and differentiated. Methods： SSCs were isolated in vitro and cultured and stem cell factors were used to induce SSCs to differentiate into sperma tocytes. The immnofluorescence staining and RT-PCR techniques were used to detect the expression of Oct-4 and GCNF before and after differentiation of mouse SSCs. Results： There existed ex- pression of Oct-4 in mouse SSCs before induction. Two days after induction, the expression decreased; the negative expres- sion of GCNF was observed before the induction of mouse SSCs, while the positive expression of GCNF was observed 2 days after induction towards spermatocyte. Conclusion. GCNF is likely to prompt the differentication of SSCs by inhibiting the expression of Oct-4.
