摘 要:目的:通过观察山茶籽对未成熟雌性大鼠乳腺、子宫发育的影响,为山茶籽治疗雌激素缺乏性疾病提供实验依据。方法:21-26d健康雌性SD大鼠,分成正常对照组、己烯雌酚组、山茶籽低剂量组和山茶籽高剂量组。测血清中雌二醇、孕酮、促卵泡激素含量,测量乳腺、子宫湿重、体乳腺质量比和子宫系数,子宫组织切片,光镜观察其形态结构。结果:己烯雌酚组和山茶籽高剂量组的体乳腺质量比和子宫系数增大,雌二醇和孕酮含量升高,而血清促卵泡激素含量降低;与山茶籽低剂量组比较,山茶籽高剂量组乳腺体质量比和子宫系数明显增大。结论:山茶籽醇提物有雌激素作用,可促进未成熟雌性大鼠乳腺和子宫生长发育及雌、孕激素分泌,山茶籽的雌激素生物活性与其剂量呈正相关。Objective: To explore the effects of Camellia seed on the development of the mammary gland and uterus in rat, in order to provide experimental basis for estrogen deficiency diseases treated with Camellia seed. Methods: Healthy female 813 rats aged from 21 to 26 days were randomly divided into four groups (n=20) according to different treatment, including nor- mal control, diethylstilbestrol group, low dose Camellia seed group and high dose Camellia seed group. Twelve days later, the levels of estradiol (E2), progesterone (P) and follicle stimulating hormone (laSH) in serum were detected, and wet weight of the mammary gland and uterus, as well as the mass ratio of body vs mammary gland and index of uterus were measured. Subsequently, the morphology of the rat uterus was observed under optical microscope. Results: Application of diethylstilbestrol and high dose Camellia seed caused increases in mass ratios of body vs mammary gland, and index of the u- terus, as well as increases in E2 and P in blood serum, while the FSH in blood serum were decreased. Compared with those in the low dose Camellia seed group, high dose Camellia seed caused obvious increases in mass ratio of body vs mammary gland and index of the uterus. Conclusion: Ethanol extract of Camellia seed has an effect comparable to estrogen, therefore, to promote the development of the immatured mammary gland and uterus of rats, and secretion of estrogen and progestogen in a dose-dependent manner.
