舌癌永生化细胞系细胞淋巴道转移裸鼠模型的建立

摘　　要：目的：建立人舌癌细胞Tca8113-M1裸鼠淋巴结转移模型，研究分析其转移的特性，为舌癌转移的研究提供动物模型。方法：4～6周龄裸鼠随机分为生理盐水对照组、Tca8113-M1组、Tca8113组，通过足垫注射舌癌细胞，建立淋巴道转移模型。观察各组裸鼠淋巴结转移情况，进行H-E染色、上皮膜抗原（EMA）免疫组织化学显色及淋巴结舌癌细胞纯化培养。结果：淋巴结有散在或灶性鳞状上皮细胞转移，体外纯化培养的癌细胞呈上皮样生长。Tca8113-M1组淋巴结转移率为88％，Tca8113组仅为60％。结论：足垫注射舌癌永生化细胞系Tca8113-M1细胞系细胞建立淋巴道转移模型具有简单、稳定、转移率高等特点，为舌癌淋巴道转移的机制研究提供了一个良好动物模型。Objective. To establish a nude mice model of lymphatic metastasis using immortalized human tongue carcinoma cell line. Methods.. Lymphatic metastasis nude mice models were established by injecting carcinoma cells into the nail pad. The lymph nodes were collected 4 weeks later. The detection methods included H-E staining, immunohistochemistry stain of EMA and the purification and culture of tongue squamous cell carcinoma cells in vitro. Results： The lymphatic metastasis rate was 88% in Tca8113-M1 group and 60% in Tca8113 group. There were lymphatic metastasis carcinomal cells in the lymph node of the tongue carcinoma metastasis model In purification and culture, the cells of epithelium like growth were observed in vitro. Conclusion： Injecting Tca8113-M1 cells into the nail pad can readily and stably establish a high rate of the lymphatic metastasis in an animal model. The model was regarded as a basis study of the lymphatic metastasis mechanisms.
