摘 要:目的研究大黄利胆胶囊对大鼠酒精性脂肪肝的影响。方法采用生理方式喂食酒精建立AFL大鼠模型,计算各组大鼠肝指数,进行肝功检测及肝组织病理学检查;生化分析法检测肝组织TG与MDA含量、抗氧化酶及ADH活性;ELISA检测血浆TNF-α含量,免疫组织化学检测肝脏CYP2E1蛋白表达。结果与模型组比较,大黄利胆胶囊低、中、高剂量组大鼠的肝指数、血清AST与ALT水平均明显降低(P<0.01或P<0.05);肝组织病理学改变减轻;TG与MDA含量均明显降低(P<0.05或P<0.01);GSH-PX均明显增加(P<0.01),SOD活性轻度增加(P>0.05);CYP2E1蛋白表达均明显下调(P<0.01);而大黄利胆胶囊中、高剂量组ADH活性明显减低及高剂量组TNF-α减少(P<0.01)。结论大黄利胆胶囊对AFL有显著的治疗作用,其作用机理可能与减轻肝脏脂肪堆积、加速乙醇清除及提高肝脏抗氧化与抗炎能力相关。Objective To investigate the effects of rhubarb choleretic capsule(RCC) on alcoholic fatty liver in rats. Methods The male SD rats with alcoholic fatty liver(AFL) were prepared by a physiological feeding the mixture of alcohol. Ten rats drunk with water without alcohol and sugar were used as normal controls. The hepatic index(HI) ,liver function and histopathological ex- amination were observed. The contents of TG and MDA and the activities of antioxidant enzyme and ADH in the hepatic tissues were measured by biochemical analysis. The plasma levels of TNF-c~ and protein expression of CYP2E1 in liver were detected by ELISA and immunohistochemistry, respectively. Results Compared with the model group, a significant reduction was observed in HI,serum ALT(U/L) and AST(U/L) in rats treated with RCC at the indicated doses (all P 〈 0.01 or P 〈 0.05 );The his- topathological changes were significantly alleviated ; The contents of TG and MDA were significantly decreased ( all P 〈 0.05 or P 〈0.01 ) ;GSH-PX activity was significantly increased (all P 〈 0.01 ) while SOD activity was slightly increased (P 〉 0.05 ) ; The protein expression of CYP2E1 were also significantly lower ( all P 〈 0.01 ). In addition, the ADH activities in the middle- and high- dose groups and the TNF-ot level high- dose group were also significantly decreased, respectively( P 〈 0.01 ). Conclusion RCC can significantly protect rats against the fatty liver damage induced by drinking alcohol,which pharmacological mechanisms were involved in the reduction of hepatic steatosis, acceleration of alcohol elimination and enhancement of anti-oxidation and anti- inflammation.
