摘  要:目的通过针刺的方法治疗阿尔茨海默病模型大鼠,观察对其学习和记忆的改善效果。方法通过给大鼠灌服D-半乳糖制剂,造成大鼠阿尔茨海默病模型。分别设立正常组、模型组、药物组、针刺组、电针组。正常组、模型组只给予相同剂量的生理盐水灌服,药物组给予石杉碱甲片(哈伯因)混悬液灌服治疗,针刺组采用针刺百会、神庭、肾俞、太溪、足三里的方法进行治疗,电针组采用针刺百会、神庭、肾俞、太溪、足三里穴,并连接电针仪进行治疗。以10 d为1个疗程,共治疗3个疗程。通过大鼠跳台试验来检测大鼠的学习和记忆能力,并对大鼠大脑海马CA1区的tau蛋白含量进行观察。结果造模后,正常组大鼠的记忆逃避潜伏期时间明显少于模型组(P<0.05),治疗结束后药物组、针刺组、电针组的逃避潜伏时间短于模型组(P<0.05)。电针组的疗效优于针刺组和药物组(P<0.05)。而治疗后,海马CA1区的P-tau蛋白表达治疗组都低于模型组,且电针组的效果要优于药物组和针刺组(P<0.05)。结论运用电针能改善阿尔茨海默病模型大鼠的记忆能力,对阿尔茨海默病有一定的治疗作用。Objective To investigate the improving effect of acupuncture treatment on rat learning and memory in a rat model of Alzheimer's disease. Method A rat model of Alzheimer's disease was made by gavage of D-galactose. Normal, model, medication, acupuncture and electroacupuncture groups were set up. The normal and model groups were given gavage of the same amount of normal saline. The medication group was treated by gavage of Huperzine A suspension; the acupuncture group, by needling points Baihui (GV20), Shenting (GV24), Shenshu (BL23), Taixi (KI3) And Zusanli (ST36); the electroacupuncture group, by needling points Baihui (GV20), Shenting (GV24), Shenshu (BL23), Taixi (KI3) and Zusanli (ST36) and providing electrical stimulation to these points. Treatment was given 10 days as a course, for a total of 3 courses. Rat learning and memory were assessed by a step down test. The tau protein content of rat hippoeampal CA1 region was determined. Result After model making, rat memory escape latency time was significantly shorter in the normal group than in the model group (P〈0.05). After the end of treatment, rat memory escape latency time was shorter in the medication, acupuncture and electroacupuncture groups than in the model group (P〈0.05). The therapeutic effect was better in the electroacupuncture group than in the acupuncture and medication groups (P〈0.05). After treatment, the expression of tau protein in the hippocampal CA1 region was lower in the treatment groups than in the model group and the effect was better in the electroacupuncture group than in the medication and acupuncture groups (P〈0.05). Conclusion Electroacupuncture can improve the memory of rat model of Alzheimer's disease, and has a certain treatment effect for Alzheimer's disease.
