海藻、甘草单用及配伍不同比例对小鼠急性毒性的影响

摘　　要：目的通过海藻-甘草不同配伍比例对小鼠急性毒性的影响,评价反药组合前后的毒性表征及其特点。方法以小鼠急性死亡率为指标,通过海藻水煎剂经口给药考察其急性毒性,计算海藻小鼠半数致死量(LD50)及95%置信限。根据海藻LD50值,固定海藻用量,结合中国药典海藻和甘草用量范围,设定海藻-甘草的5个比例:1∶3、1∶2、1∶1、1∶0.33、1∶0.17。同时,设甘草最大用量组90g/kg和海藻30g/kg单药对照组,观察动物的毒性反应症状和死亡情况,连续观察14天,进行解剖和观察结果分析。结果海藻-甘草比例为1∶3和1∶1时,小鼠死亡率为100%;比例为1∶2时,死亡率为80%;海藻单药组和比例为1∶0.33时,死亡率为60%;比例为1∶0.17时,死亡率为50%;甘草单药组的死亡率为0。结论药组中甘草大于等于海藻时,表现出增毒效应。OBJECTIVE The present study has evaluated the toxicity of different compatibility ratio of Sargassum pallidum（Turn.）C.Ag.and Glycyrrhiza uralensis Fisch.By determining mortality after single oral administration in ICR mice.METHODS The median lethal death（LD50） and its 95% confidence limit were calculated by the morality of mice in acute toxicity testing.Five different compatibility ratios 1∶3,1∶2,1∶1,1∶0.33,1∶0.17 of Sargassum and Glycyrrhiza were designed in accordance with the dosage range of Sargassum and Glycyrrhiza in the Pharmacopeia,in which the dose of Sargassum was fixed on the basis of the LD50.Otherwise,the high dose 90 g/kg of Glycyrrhiza and the dose 30 g/kg of Sargassum are setted as two control groups.General behavior,adverse effects and mortality were recorded for up to 14 days post treatment.RESULTS When the ratio of Sargassum and Glycyrrhiza is 1∶3 and 1∶1,the mice morality is 100%;the ratio of two herbs is 1∶2,the mice morality is 80%;the Sargassum control group and the ratio of two herbs is 1∶0.33,the morality is 60%;the ratio is 1∶0.17,the morality is 50%;the morality of Glycyrrhiza control group is 0%.CONCLUSION The results show that toxic effects are increased when the dose of Glycyrrhiza is higher than the dose of Sargassum.
