益气温阳方对心力衰竭大鼠心脏血流动力学及心肌能量代谢的影响  

Effect of "Yiqi Wenyang Prescription" on Cardiac Hemodynamics and Myocardial Energy Metabolism in Rats with Heart Failure

摘　　要：目的:观察益气温阳方对冠状动脉结扎致充血性心力衰竭大鼠心脏血流动力学及心肌能量代谢的影响。方法:将60只SD大鼠随机分为假手术组、模型组、曲美他嗪对照组及益气温阳方高、中、低剂量组,通过冠状动脉结扎法制备充血性心力衰竭大鼠模型。药物灌胃干预4周后采用电生理仪记录血流动力学变化,高压液相色谱法测定心肌细胞中ATP、ADP、AMP含量变化。结果:益气温阳方组心率、左室内压最大上升和下降速率(±dp/dt max)均高于模型组(P<0.01),左室舒张末压(LVEDP)低于模型组(P<0.01),益气温阳方高、中剂量组略优于曲美他嗪组;益气温阳方组大鼠心肌组织ATP含量显著高于模型组(P<0.01),ADP、AMP含量显著低于模型组(P<0.01),三组疗效略差于曲美他嗪组。结论:益气温阳方可以有效改善心肌梗死后心力衰竭大鼠模型心脏功能,同时提高心衰大鼠心肌能量代谢产物ATP含量,减少AMP、ADP的含量,有效调节心肌梗死后心衰大鼠心肌能量代谢紊乱。Objective： To investigate the effects of ＂Yiqi Weuyang Prescription＂ on cardiac hemodyuamics and myocardial energy metabolism in rats with congestive heart failure （CHF） induced by coronary artery ligation. Methods： The CHF models were efftablished by coronary artery ligation. Sixty rats were randomly divided into sham-operated group, model group, Trimetazidine group and ＂ Yiqi Wenyang Prescrition＂ groups （large, medium, small dose ）. After 4 weeks of intragastric administration, the hemodynamic diversity was recorded by electro-physiograph, the contents of ATP, ADP and AMP were measured by high pressure liquid chromatography. Results： The heart rate, ＋ dP/dtmax and -dP/dtmax in ＂Yiqi Wenyang Prescrition＂ groups were higher than those in model group （P 〈0.01 ）, and the LVEDP was lower than that in model group （P 〈0. 0l ）. The efficacy of ＂Yiqi Wenyang Prescrition＂ groups of large and medium dose was better than the trimetazidine group; the content of ATP in cardiac muscular tissue of ＂Yiqi Wenyang Prescrition＂ groups was higher than that in model group markedly （ P 〈0.01 ）, the contents of ADP and AMP were lower than that in model group （ P 〈0.01 ）. The efficacy of three ＂Yiqi Wenyang Prescrition＂ groups Was worse than the trimetazidine group. Conclusion： ＂Yiqi Wenyang Prescription＂ can improve cardiac function, increase the content of ATP, decrease the contents of AMP and ADP, and regulate the energy metabolism of heart failure after myocardial infarction in rats.
