摘 要:目的探讨七叶灵颗粒在抑制肿瘤生长及肿瘤细胞侵袭和转移方面的作用机制。方法将接种的C57小鼠随机分为4组:不用任何治疗的模型组(A组)、化疗组(B组)、七叶灵颗粒组(C组)和化疗加七叶灵颗粒组(D组),观察各组小鼠生存质量,比较各组小鼠的体内抑瘤作用。采用原位细胞凋亡检测试剂盒检测标本组织凋亡细胞数,用原位杂交检测,检测标本组织MMP9阳性表达数。结果①C组和D组小鼠的生存质量优于A组和B组。②D组的抑瘤率为74.47%,显著高于A组(P<0.01)。B、C、D组的平均瘤重显著低于A组(P均<0.05)。③原位细胞凋亡检测表明,B组、C组和D组瘤组织凋亡细胞数均显著高于A组(P均<0.05),且D组显著高于C组(P<0.05)。④原位杂交检测表明,D组、C组和B组肺癌组织中的MMP9表达水平均显著低于A组(P均<0.05)。结论七叶灵颗粒有一定的改善C57荷瘤小鼠生存质量、抑制Lewis肺癌细胞生长、诱导Lewis肺癌细胞凋亡及抑制MMP9活性表达的作用。Objective To observe the effects of "Qiyeling Granule" in suppressing tumor growth, and infiltration and metastasis of tumor cells of C57 mice with transplanted tumor. Methods C57 mice with transplanted tumor were randomized into four groups : model group ( group A), chemotherapy group ( group B), "Qiyeling Granule Group" group( group C) and chemotherapy plus "Qiyeling Granule Group" group (group D). The living quality and suppression of tumor growth of C57 mice were observed ; the in situ cell apoptosis analysis kit was used for count of apoptosis, and hybridisation was used to test MMP9 expression. Re- suits ①Quality of life was better in groups C and D than in groups A and B. ②Suppression rate of tumor growth was 74.47% in group D, obviously higher than that in group A( P 〈 O. O1 ), and the average tumor weight was lower in group B, C and D than in group A( all P 〈 0.05 ). ③In situ cell apoptosis analysis showed that tumor apoptosis was higher in group B, C and D than in group A ( P all 〈 0.05 ), higher in group D than in group C ( all P 〈 0.05 ). ④In situ hybridisation test indicated that MMP9 expressions were significantly lower in group B, C and D than in group A ( all P 〈 O. 05 ). Conclusions "Qiyeling Granule" can im- prove the living quality of C57 mice, and suppress the growth of Lewis lung cancer cell by inducing apoptosis of cancer cells and inhibiting MMP9 expression.
