摘 要:目的观察参龙汤对脑缺血大鼠的脑保护作用及对血管内皮生长因子(VEGF)表达的影响。方法利用双侧颈总动脉结扎的方法制备慢性脑缺血模型,50只大鼠分为假手术组、模型组、脑复康组、脑缺血加参龙汤大、小剂量组。造模以及药物处理4周后,进行各组大鼠神经功能缺失状态评分及大鼠海马区VEGF阳性细胞表达检测。结果模型组大鼠神经功能缺失症状明显,与假手术组比有显著差异。参龙汤能改善脑缺血大鼠神经功能缺失症状,参龙汤大剂量组与模型组相比有显著差异。模型组大鼠海马区VEGF阳性细胞表达明显减少,参龙汤大剂量治疗组大鼠海马VEGF阳性细胞表达增加,与模型组比面积大、颜色深,平均光密度值与模型组比差异有统计学意义。结论参龙汤对双侧颈总动脉结扎脑缺血大鼠有脑保护作用,能改善大鼠神经功能缺失症状,其作用机理与增加VEGF的表达有关。Objective: To explore the protective effects of Shenlongtang(SLT) on neuron protection and the VEGF expression on rats with cerebral ischemia model.Methods: The cerebral ischemia animal model was established by two-vessel occlusion.50 rats were randomly divided into control group with slam operation,model group,ischemia model with positive control group of pisacetam,ischemia model with SLT low and high dosage groups.Four weeks administrated with drug,the neuron logic function was observed and expression of VEGF in hippocampus was detected.Result: Compared with sham operation group,model group presented severe neuron logic deficits and significantly increases of VEGF expression in hippocampus.Compared with ischemia model group,neuron logic deficits in SLT high dosage groups was relieved significantly and VEGF expression increased distinctly in SLT groups.Conclusion: SLT could relive cerebral ischemia injury in rats by increasing VEGF expression in hippocampus.
