摘　　要：应用高效液相色谱法(HPLC)测定犬血浆中伊曲康唑的浓度。选用6条健康犬,以4 mg/kg体重背部皮下注射给药,采用HPLC测定犬血浆中伊曲康唑浓度(ITZ),应用3p97软件处理数据,计算药代动力学参数。结果表明血浆ITZ线性范围为10～1 000 ng/mL,伊曲康唑血浆添加浓度10、100、500ng/mL的平均回收率分别为86.9%、88.4%、89.2%,RSD分别为2.8%,1.2%,0.2%。说明HPLC法简便,准确可靠,适用于伊曲康唑在犬体药动学中血药浓度测定。Pharmacokinetics ofitraconazole injection was studied in healthy dogs. Subcutaneous injection was administrated to six dogs in a dose of 4mg/kg and the plasma concentration of itraconazole was detected by HPLC. The data was analyzed with 3p97 computer program. The results showed that the linearity was obtained over the range of 10-1000ng/mL and the recovery was 86.9%, 88.4% or 89.2% when the addition concentration of itraconazole was 10 ng/mL/, 100 ng/mL or 500 ng/mL respectively. The RSD was 2.8% , 1.2% or 0.2% respectively. It was concluded that the method was simple, accurate and reliable, and could be used for the pharmacokinetic studies ofitraconazole in dogs.
