鸦胆子油乳注射液联合普通放疗对C6胶质瘤细胞作用的实验研究

摘　　要：目的探讨鸦胆子油乳注射液联合普通放疗对C6胶质瘤细胞增殖的影响。方法体外培养大鼠C6胶质瘤细胞,用MTT比色法检测鸦胆子油乳注射液联合普通放疗对C6胶质瘤细胞的抑制作用。用免疫组化检测鸦胆子油乳注射液联合普通放疗对C6胶质瘤细胞caspase-3表达的影响。结果 MTT比色法显示鸦胆子油乳注射液联合放疗对C6胶质瘤细胞抑制率高于单次普通放疗;免疫组化显示鸦胆子油乳注射液联合放疗使C6胶质瘤caspase-3表达效果高于单次普通放疗。药物作用呈剂量依赖性。普通放疗联合鸦胆子油乳注射液与单纯用药相比对肿瘤细胞抑制率及肿瘤细胞caspase-3表达无明显差异(P>0.05)。用药后行普通放疗对C6胶质瘤细胞的抑制率及细胞caspase-3表达低于用药前行放疗。结论体外鸦胆子油乳注射液能增加C6胶质瘤细胞凋亡基因caspase-3表达,抑制细胞增殖。鸦胆子油乳注射液联合普通放疗对C6胶质瘤细胞抑制作用高于普通放疗。药物联合放疗与单独用药对C6胶质瘤细胞作用无明显差异。放疗后用药优于放疗前用药。Objective To explore the effect of brucea javanica oil emulsion combined with conventional radiotherapy on proliferation of C6 glioma cells.Methods Rat C6 glioma cells were cultured in vitro.MTT colorimetric assay was used to measure the inhibitory effect of brucea javanica oil emulsion combined with conventional radiotherapy on C6 golima cells.Immunohistochemical staining was used to detect the effect of Brucea javanica oil emulsion combined with conventional radiotherapy on the caspase-3 expression of C6 glioma cells.Results MTT demonstrated that the inhibition rate of Brucea javanica oil emulsion combined with conventional radiotherapy on C6 glioma cells was higher than radiotherapy alone;the immunohistochemistry showed that the effect of Brucea javanica oil emulsion combined with conventional radiotherapy on the caspase-3 expression of C6 glioma cells was higher than radiotherapy alone too.The effect of drug was does-dependent.Compared brucea javanica oil emulsion alone with conventional radiotherapy combined with brucea javanica oil emulsion,there were no significant differences in inhibition rate on C6 glioma cell and caspase-3 expression of C6 glioma cell.The inhibition rate on C6 glioma cell and caspase-3 expression of C6 glioma cell in groups of drug after radiotherapy were higher than those in groups of drug before radiotherapy.Conclusion Brucea javanica oil emulsion can inhibit proliferation of C6 glioma cells by increasing the expression of caspase-3 in vitro.The inhibitory effect of brucea javanica oil emulsion combined with conventional radiotherapy on C6 glioma cells is higher than radiotherapy alone.There are no significant differences between the effects of brucea javanica oil emulsion alone and brucea javanica oil emulsion combined with radiotherapy on C6 glioma cells.The effect of drug after radiotherapy is better than it of drug before radiotherapy.
