摘 要:目的探讨中药复方对小鼠吗啡镇痛耐受的影响及其机制。方法选取2008年12月—2009年5月健康雄性昆明种小鼠30只,随机数字表法分为3组,即吗啡组(M组)、吗啡加中药复方组(MB组)、正常对照组,每组10只。在安静、室温、正常光照的环境中适应性饲养1周,1周后M组每12 h皮下注射吗啡10 mg/kg;MB组每12 h皮下注射吗啡10 mg/kg并给予中药复方0.2 ml/只。以上各组均给药1周共15次。正常对照组不给予任何处理。第1次给药前及第1、3、6、9、12、15次给药半小时后测量甩尾潜伏期(TFL)。免疫组化染色测定蛋白激酶A(PKA)。结果 M组与MB组给药前、第1次、第3次给药后TFL比较,差异均无统计学意义(P>0.05);两组第6次、第9次、第12次、第15次给药后TFL比较,差异均有统计学意义(P<0.05)。正常小鼠脑皮质免疫组化染色显示PKA几乎没有表达,M组PKA表达有所增多,MB组小鼠脑皮质内PKA明显增多。M组PKA染色平均光密度值为(0.3492±0.0055),MB组为(0.3230±0.0096),差异有统计学意义(t=2.83,P<0.05)。结论中药复方对小鼠吗啡耐受的形成具有抑制作用,与吗啡同用可以取得良好的镇痛效果,其机制可能与拮抗了吗啡对细胞内信号转导系统的影响有关。Objective To observe the effect of Traditional Chinese Medicinal Compound on the development of morphine tolerance in rats and its mechanism. Methods 30 male km mice from December 2008 to May 2009 were randomly divided into three groups (M group, MB group and control group) with each group 10 mice. All the mice were bred adaptively in a quiet environment with room temperature and normal illumination for one week. And then the mice in the M group were given subcutaneous injection of morphine for 10 mg/kg every 12 h. The mice in the MB group were given subcutaneous injection of morphine for 10 mg/kg as well as Traditional Chinese Medicinal Compound for 0. 2 ml/mouse. All the mice were treated 15 times for one week. The control group was not given any treatment. TFL was measured before drug administration and 30 minutes after the 1st, 3rd, 6th, 9th, 12th and 15th drug administration respectively. Immunohistochemical method was used to detect PKA. Results The TFL between the two groups showed no statistically significant differences before drug administration and after the 1 st and 3rd drug administration (P 〉0. 05). While the TFL between the two groups showed statistically significant differences after the 6th, 9th, 12th and 15th drug administration (P 〈0. 05) . PKA had little expression in cerebral cortex among normal mice according to immuno- histochemistry. While the expression of PKA increased in the M group and increased significantly in the MB group. The average OD value of the M group and MB group was (0. 3492 ± 0. 0055 ) and (0. 3230 ± 0. 0096) respectively ( t = 2. 83, P 〈 0.05 ). Conclusion The Traditional Chinese Medicinal Compound can inhibit the development of morphine tolerance in rats and can achieve satisfactory analgesic effect when combined with morphine. Its mechanism may relate to the antagonism on the influence of signal transduction system by morphine.
