摘  要:采用响应面优化法研究肿节风多糖的超声提取工艺及纯化得到的肿节风多糖(SGP-U)对人肺癌细胞株(NCI-H460)的抑制作用。选择提取时间、超声功率、液料比作为影响因素,以多糖提取率为评价指标,进行单因素考察。在此基础上,应用Box-Behnken中心法则,经响应面(RSM)软件分析得到最优提取条件。超声提取多糖经纯化后得到肿节风多糖SGP-U,并采用四甲基偶氮唑盐(MTT)比色法检测其对NCI-H460细胞的抑制作用。在最佳优化条件为提取时间46 min、超声功率180 W、液料比44 mL/g时,多糖提取率理论值为6.35%,验证值为(6.31±0.09)%。利用响应面优化法可以获得肿节风多糖超声提取的最优工艺条件,并且经纯化后的SGP-U对NCI-H460肿瘤细胞具有抑制作用。To study on the ultrasonic-assisted extraction of polysaccharides (SGP) from Sarcandra glabra (Thunb.) Nakail and the inhibition of SGP-U on the proliferation of human lung cancer cell line (NCI-H460), response surface methodology (RSM) was used to optimize the ultrasonic-assisted extraction parameters. Box-Behnken central-composite design ( BBD ) was applied to estimate the effects of extraction time, ultrasonic power and liquid/solid ratio on the yield of SGP. The inhibition of purified SGP (SGP-U) on the proliferation of human lung cancer cell line was determined by [ 3-( 4,5-dimethyhhiazol-2-yl ) -2,5-diphenylte-trazolium bromide ] MTT assay. Optimum extraction conditions were determined as follows: extraction time of 46 min, uhrasonic power of 180 W, and liquid/solid ratio of 44 mL/g. Under these conditions, the estimated and observed values of maximal yield of polysaccharides were 6. 35% and (6. 31 ± 0. 09) % , respectively. The optimum ultrasonic extraction conditions were obtained by RSM, and SGP-U could inhibit the proliferation of human lung cancer cell (NCI-H460).
