六味地黄生物制剂对衰老模型小鼠脑海马区神经元Bcl-2和Bax mRNA表达的影响  被引量:5

摘  要:目的:观察六味地黄生物制剂(LW-PSB)对衰老模型小鼠脑海马区神经元Bcl-2和Bax mRNA表达的影响。方法:将昆明种小鼠随机分为模型组、阳性对照维生素E(VE 0.27 g.kg-1)组、LW-PSB治疗组(0.50 g.kg-1)和正常对照组,共4组,每组各10只。造模后第15天开始按0.02 mL.g-1体重灌胃给药,模型组、正常对照组灌胃等体积生理盐水,每日1次,连续灌胃30 d。末次给药30 min后处死并分别取各组小鼠脑海马体,采用RT-PCR法检测各组小鼠脑海马区神经元Bcl-2和Bax mRNA的表达。结果:与模型组比较,LW-PSB治疗组Bcl-2 2-△△Ct显著升高(P<0.01),阳性对照组和LW-PSB治疗组Bax 2-△△Ct显著降低(P<0.01),阳性对照组和LW-PSB治疗组Bcl-2/Bax比值显著升高(P<0.01);与阳性对照组比较,LW-PSB治疗组Bcl-2 2-△△Ct升高(P<0.05)、Bax 2-△△Ct降低(P<0.05)和Bcl-2/Bax比值显著升高(P<0.01)。结论:六味地黄生物制剂通过促进脑海马区神经元Bcl-2 mRNA的表达和抑制Bax mRNA的表达起到抗衰老作用。Objective: To investigate the effect of Liuwei Dihuang decoction metabolized by photosynthetic bacteria (LW-PSB) on .the expression of Bcl-2 and Bax mRNA in hippocampus neurons of aging model mice. Method : Mice were randomly divided into 4 groups : model group, positive control group (rE 0.27 g ·kg^(-1) ) , LW-PSB-treated group (0.50 g·kg^(-1) ) and normal group. Each group was administrated continuously for 30 days. After 30 rain of administration, the hippocampus of each group was separated and RT-PCR was adopted to detect the expression of Bcl-2 and Bax mRNA in hippocampus neurons. Result: Compared with the model group and positive control group, the Bcl-2 2^(-△△c1) and Bcl-2/Bax were increased and Bax 2^(-△△c1)was decreased in LW-PSB-treated group. Conclusion: LW-PSB might up-regulate expression of Bcl-2 mRNA and down-regulate expression of Bax mRNA to anti-aging.
