摘 要:目的:探讨毒素清颗粒对支原体肺炎老龄大鼠免疫功能和细胞因子的影响。方法:SD大鼠随机分为青年对照组、青年模型组,老龄对照组、老龄模型组、毒素清组和阿奇霉素组,采用气管插管法复制支原体肺炎模型。分别以毒素清(3.995 g.kg-1.d-1)、阿奇霉素(23 mg.kg-1.d-1)灌胃7 d。第8天取材,检测外周血白细胞(WBC)和中性粒细胞(NEU)计数,支气管肺泡灌洗液(BALF)支原体PCR和分泌性免疫球蛋白(sIg)A、纤维连接蛋白(FN)表达,肺脏含水量和病理,血清肿瘤坏死因子(TNF)-α、白介素(IL)-6、IL-8表达。结果:阿奇霉素组BALF肺炎支原体阳性为0例(0%),毒素清组阳性3例(30%)。青年和老龄模型组BALF中sIgA和FN分别较青年、老龄对照组显著降低(P<0.01),肺组织含水量、外周血TNF-α,IL-6,IL-8显著升高(P<0.01),且较青年模型组变化显著(P<0.01)。与老龄模型组比较,毒素清组和阿奇霉素组sIgA,FN显著升高(P<0.01),肺组织含水量、TNF-α,IL-6,IL-8显著降低(P<0.01);毒素清组BALF中sIgA明显高于阿奇霉素组(P<0.01),IL-8低于阿奇霉素组(P<0.05)。结论:毒素清对老龄大鼠支原体肺炎作用明显,其机制可能与增强免疫功能、降低炎症因子表达,减轻炎症反应有关。Objective: To explore the effect of Dusuqing granule on immune function and cytokines in the aged rat with mycoplasma pneumonia (MP). Method: Young rats were randomly divided into young control and young model groups, aged rats into aged control, aged model, Dusuqing and Azithromycin groups. Mycoplasma pneumonia rat model was duplicated by trachea intubation, and were ig administrated with Dusuqing granule (3. 995 g ·kg^(-1)·d^(-1)) and Azithromycin (23 mg ·kg^(-1)·d^(-1)) for 7 days, and sacrificed at day 8. White blood cells (WBC) and neutrophilie granulocyte (NEU) count in peripheral blood were observed, and mycoplasma polymerase chain reaction (PCR), levels of secretory immunoglobulin (sIg) A and fibronectin (FN) in bronchoalveolar lavage fluid (BALF), lung water content and pathological changes, and expressions of tumor necrosis factor (TNF)-a, interleukin (IL)-6, IL-8 in serum were also measured. Result: Mycoplasma is negative in Azithromycin group (0 animal/0% ), positive in Dusuqing group (3 animals/30% ). Compared to young and aged control group, levels of sIgA and FN in BALF significantly decreased in young and aged model group (P 〈 O. 01), and expressions of TNF-a, IL-6 and IL-8 increased in serum (P 〈 0.01 ), respectively. And those changed obviously in aged model group than in young model group (P 〈 0.01 ). Compared to aged model group, levels of slgA and FN significantly increased in Dusuqing and Azithromycin groups ( P 〈 0.01 ) , while lung tissue water content, TNF-a, IL-6 and IL-8 decreased (P 〈0. 01). And sIgA Azithromycin group (P 〈 0.01), and IL-8 was lower (P 〈 0. 05).
