摘 要:目的通过不同方式建立大鼠血管性ED模型,并对模型结果进行验证,比较各种造模方法的有效率,并探讨模型的稳定性。方法将75只雄性SD大鼠随机分为单纯手术组(18只,双侧髂内动脉结扎)、单纯高脂组(20只,高脂饲养)、手术+高脂组(25只,双侧髂内动脉结扎,高脂饲养)、假手术组(4只,未行血管结扎术)、正常对照组(8只)。术后6周,通过交配试验进行动物行为学观察,通过阿朴吗啡(APO)诱导下进行阴茎海绵体内压连续监测,记录最大勃起的海绵体内压改变,客观评价勃起功能。结果在交配试验中,手术±高脂组均没有出现交配行为(爬高、插入);单纯手术组有3例出现爬高现象,但均无插入;高脂组有8例出现爬高现象,其中2例有插入现象。海绵体测压结果显示:正常对照组(32.89±6.42)mmHg,手术+高脂组(3.21±1.20)mmHg,单纯手术组(9.62±3.87)mmHg,单纯高脂组(12.42±5.11)mmHg,假手术组(29.48±4.83)mmHg。结论手术+高脂是目前建立血管性ED的最有效方法,且模型稳定。Objectives To establish different rat vascular ED models and investigate the stability of the model Methods Total of 75 male SD rats were randomly divided into five groups including the surgery group (18 rats, bilateral internal iliac artery ligation), the high-fat group (20 rats, high-fat diet), the surgery and high-fat group (25 rats, iliac artery ligation, high-fat diet), sham operation group (4 rats, no vascular ligation) and the normal control group (8). Six weeks postoperatively, the animal behavior was evaluated by the mating test. Intracavernous pressure induced by apomorphine (APO) was assessed continuously. The biggest erection ofintracavernous pressure change was recorded for evaluation of the erectile function. Results The mating behavior (climb, insert) was not found in the surgery and high-fat group. Climb phenomenon was found in three cases of the surgery group, but no insert phenomenon was found. Climb phenomenon was observed in eight cases of the high-fat group, and insert phenomenon was found in two cases of them. The average intracavernous pressures of the normal control group, the surgery and high-fat group, the surgery group, the high-fat group and sham operation group were (32.89 ±6.42)mmHg, (3.21 ±1.20)mmHg, (9.62 ± 3.87)mmHg, (12.42±5.11)mmHg, and (29.48 ± 4.83)mmHg respectively. Conclusion Surgery combining with high-fat was the most effective method for establishment of vascular ED model.
