摘 要:目的探讨载紫杉醇脂质体微泡(paclitaxel-carrymg liposomemicrobubbles,PLM)联合低频超声干预低龄兔髂动脉血管结构重建的可行性。方法低龄新西兰大白兔单侧髂动脉狭窄模型25只,随机平均分为5组,A组:PLM联合低频超声治疗;B组:空白微泡联合低频超声治疗;C组:紫杉醇药物联合低频超声治疗;D组:低频超声辐照治疗;E组:空白对照组。建模后3周进行干预,4周取损伤处血管,HE染色计算血管管腔面积、内膜面积及中膜面积,免疫组化分析血管壁平滑肌细胞的增殖情况。结果各干预组髂动脉狭窄模型的管腔面积均大于E组(P<0.05),内、中膜面积均小于E组(P<0.05),A组管腔面积最大,内、中膜面积最小(P=0.000),B、C、D组之间没有明显差异(P>0.05)。干预后各组内膜/中膜面积比值均减小(P<0.05),以A组减小最为显著(P=0.000)。免疫组化显示各干预组幼兔髂动脉内膜层均出现α平滑肌肌动蛋白(αSM actin)阳性反应的棕黄色颗粒,E组最多,A组表达最少(P<0.05)。结论载紫杉醇脂质体微泡联合低频超声可用于干预低龄兔髂动脉血管狭窄,减轻损伤内中膜的病理性增生。Abstract:Objective To study prevention of vascular reconstruction with paclitaxel-carrying liposome microbub- bles(PLM)combining with low-frequency ultrasound in young New Zealand rabbits. Methods 25 iliac artery stenosis modles in young New Zealand rabbits were randomly divided into 5 groups. They received different treatments. The area of lumen, lining endothelium and tunica media of the iliac artery were measured by HE dyeing and immunohisto- chemistry dyeing after 1 week of the treatment. Results The lumen area of the iliac artery in the treatment groups were greater than that in group E(P〈0.05),but the intima area and the tunica media area were lower(P〈0.05). In group A, the lumen area was the largest but the intima area and the tunica media area were the lowest(P=0. 000). It reduced the ratio of the intima area and tunica media area after the treatment(P〈0. 05). Conclusions PLM combining with low-frequency ultrasound could prevent the vascular reconstruction.
