摘 要:目的探讨不同低频诊断级超声参数联合微泡对正常肝血管通透性的影响,优化超声参数以实现肝血管高通透性,减少对正常肝细胞的影响。方法使用低频诊断超声辐照造影剂显影的肝脏,按实验分组设置不同水平的辐照时间、机械指数(mechanical index,MI)、微泡剂量,经大鼠尾静脉注入50 mg/kg伊文氏蓝(Evens blue,EB)。观察辐照后肝组织中EB渗出情况,定量检测肝组织中EB含量的改变以探索不同参数条件下肝血管通透性的变化,同时对肝脏进行组织学检查观察细胞损伤情况。结果随着辐照时间的延长、MI的升高、微泡浓度的增加,肝组织中EB含量均有明显增加;HE染色检测肝细胞无明显损伤。结论低频诊断超声联合微泡可增加肝脏血管通透性,辐照时间、机械指数、微泡浓度是其重要影响因素。Objective To investigate the effects of different parameters on liver permeability under microbubble enhanced low frequency ultrasound exposure, and to optimize treatment paraments in order to obtain best result. Meth- ods The rat livers were exposed to contrast-enhanced low frequency diagnostic ultrasound with different groups of ex- posure time,mechanical index(MI), microbubble concentration. Evens blue effusion status in liver was observed and analyzed Qualitatively and quantitatively , the EB amount was compared between groups to determinate the optimal Ul- trasonic irradiation Parameter. Results Liver permeability was increased with exposure time,MI and microbubble con- centration, while no obvious injure was observed in the liver cell. Conclusions Low-frequency diagnostic ultrasound combined with contrast agent can increase the liver vascular permeability. The exposure time, MI and microbubble concentration were important influence factors,Optimal paraments can increase liver vascular permeability to serve as a valueable tool on clinical treatment.
