摘  要:探讨了恶臭假单胞菌TS1138菌体透性化细胞的制备工艺,并对所得透性化细胞转化DL-ATC生成L-半胱氨酸的反应特性进行了研究.结果表明,以2%的甲苯为渗透剂,在25℃条件下对浓度为20%的菌悬液渗透处理30 min,所得透性化细胞的酶活达1 475 U/g,为未渗透细胞的10倍.该透性化细胞的最适转化反应温度为35℃,最适反应pH为8.0,在pH 7.5~9.0范围内稳定,Cu2+、Ag+和Zn2+对透性化细胞的转化反应具有较强抑制作用,Cu2+的抑制作用最强.The permeabilization of Pseudomonas putida TS1138 cells and the properties of perme- abilized cells involved in the conversion of DL-ATC to L-cysteine were studied. A proper process was achieved with 20% of wet cell suspension permeabilized by 2% of toluene for 30 rain at 25 ~C. The activity of permeabilized cells reached 1 475 U/g which was 10 times as much as that of rest cells. The permeahilized cells exhibited optimal conversion activity at 35 ℃and pH 8.0, and were stable from pH 7.5 to 8.0. Its conversion activity was inhibited strongly by Cu2+ and moderately by Ag + and Zn2 +.
